抗痤疮中药复方筛选及乳凝胶制剂的制备

摘要:
目的:筛选对痤疮关键致病菌具有协同抑菌作用的中药组分,并据此研制一种稳定高效的中药复方乳凝胶。方法:通过文献频次分析结合药敏实验,筛选对痤疮丙酸杆菌、金黄色葡萄球菌及表皮葡萄球菌均有显著抑制作用的中药提取物,并测定其最小抑菌浓度(minimum inhibitory concentration, MIC);通过正交实验,以抑菌圈直径为指标,优化各组分配比,并考察物理性状与稳定性,筛选并确定乳凝胶的最佳配方。结果:抑菌实验证实,黄连水提物、黄芩醇提物及黄柏醇提物为协同抑菌的核心组分,对上述3种致病菌的抑菌圈直径分别可达22.5 mm、22.0 mm、31.6 mm。正交试验确定最佳复方配比为黄连水提物32 mg/mL、黄柏醇提物1 mg/mL、黄芩醇提物32 mg/mL。以此为核心药效组分,成功构建了乳剂相(药物∶液体石蜡∶Tween 80∶丙二醇:对羟基苯甲酸甲酯=1∶7∶0.6∶7∶0.02)与凝胶相(1% Carbopol 940)以1∶1比例组成的乳凝胶体系。该制剂性状均匀,在8℃、25℃、40℃下贮存28 d,其物理状态与pH值均保持稳定。结论:本研究筛选了抗痤疮中药复方,并成功制备了一种抑菌活性显著、稳定性良好的乳凝胶,其核心配方明确、工艺简便,为开发治疗痤疮的新型外用中药制剂提供了实验依据。

Screening of compound traditional Chinese medicine prescriptions for acne and preparation of emulsion gel

Abstract:
Objective: To identify traditional Chinese medicine (TCM) components with a synergistic antibacterial effect against the key pathogenic bacteria of acne, and to develop a stable and efficient compound TCM emulsion gel. Methods: The literature frequency analysis and drug susceptibility testing were used to identify TCM extracts with a significant inhibitory effect on Propionibacterium acnes, Staphylococcus aureus, and Staphylococcus epidermidis, and their minimum inhibitory concentrations MICs were determined. With the diameter of inhibition zone as an indicator, orthogonal experiments were used to optimize the ratio of each component, and the physical properties and stability of components were examined to screen and determine the optimal formulation of the emulsion gel. Results: The antibacterial test confirmed that the water extract of Coptis chinensis, the alcohol extract of Scutellaria baicalensis, and the alcohol extract of Phellodendron chinense were the core components for synergistic antibacterial action, and the diameter of the inhibition zones against the above three pathogenic bacteria reached 22.5 mm, 22.0 mm, and 31.6 mm, respectively. The orthogonal experiment determined the optimal compound ratio of 32 mg/mL water extract of Coptis chinensis, 1 mg/mL alcohol extract of Phellodendron chinense, and 32 mg/mL alcohol extract of Scutellaria baicalensis. These extracts were used as the core active components to successfully construct an emulsion gel system composed of an emulsion phase (at a ratio of 1∶7∶0.6∶7∶0.02 for drug, liquid paraffin, Tween 80, propylene glycol, and methylparaben) and a gel phase (1% Carbopol 940) at a ratio of 1∶1. This preparation exhibited uniform properties, with stable physical state and pH value after being stored at a temperature of 8 ℃, 25 ℃, and 40 ℃ for 28 days. Conclusion: This study obtained a compound TCM prescription for acne and successfully prepared an emulsion gel with a significant antibacterial activity and good stability. This emulsion gel has a clear core formulation with a simple and convenient preparation process, providing an experimental basis for the development of a novel topical TCM preparation for treating acne.
