目的:基于正交试验设计研究人参来源囊泡的提取工艺,并在炎症细胞模型上比较不同提取工艺的人参来源囊泡的作用。方法:采用正交试验设计,计算人参来源囊泡的相对纯度(粒子浓度/蛋白浓度),分析料液比、离心时间、离心力3个因素对人参来源囊泡相对纯度的影响,优化提取工艺。构建脂多糖诱导的RAW264.7细胞炎症模型,对比不同提取工艺囊泡的抗炎活性。结果:提取条件为料液比1∶1.5、离心时间60 min、离心力100 000 g时人参来源囊泡纯度最优。在抗炎活性方面,不同提取工艺的样品对炎症因子的作用各不相同,其中样品2、3、4、5均能抑制白细胞介素-6(interleukin-6,IL-6)、趋化因子-2(C-C motif chemokine ligand,CCL-2)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)的表达,样品1对TNF-α的表达无明显抑制作用,样品6、7、8对IL-6的表达无明显抑制作用,样品9仅能抑制TNF-α的表达。结论:不同提取工艺影响人参来源囊泡纯度和其抗炎效果,本研究为建立人参来源囊泡的提取工艺标准和其抗炎活性研究提供了参考。 |

Optimization of the extraction process for ginseng-derived vesicles and validation of their in vitro anti-inflammatory activity |

Objective:To investigate the extraction process of ginseng-derived vesicles using the orthogonal test,and to compare the effect of vesicles obtained by different extraction processes using an inflammatory cell model.Methods:The orthogonal test was performed to calculate the relative purity (particle concentration/protein concentration) of ginseng-derived vesicles,and the impact of the three factors of solid-liquid ratio,centrifugation time,and centrifugal force on the relative purity of ginseng-derived vesicles were analyzed to optimize the extraction process.RAW264.7 cells were used to establish a cell model of lipopolysaccharide-induced inflammation,and anti-inflammatory activity was compared between the vesicles obtained from different extraction processes.Results:The optimal extraction parameters for ginseng-derived vesicles were a solid-liquid ratio of 1:1.5,a centrifugation time of 60 minutes,and a centrifugal force of 100 000 g.As for anti-inflammatory activity,the samples obtained from different extraction processes had different inhibitory effects on inflammatory cytokines:samples 2,3,4,and 5 could inhibit the expression of interleukin-6 (IL-6),C-C motif chemokine ligand 2,and tumor necrosis factor-α (TNF-α);sample 1 had no significant inhibitory effect on the expression of TNF-α;samples 6,7,and 8 had no significant inhibitory effect on the expression of IL-6;sample 9 could only inhibit the expression of TNF-α.Conclusion:Different extraction processes influence both the purity and anti-inflammatory activity of ginseng-derived vesicles,and this study provides a reference for establishing a standardized extraction process for ginseng-derived vesicles and conducting the research on their anti-inflammatory activity. |
