基于肠道菌群探讨蜂针疗法对佐剂性关节炎大鼠的干预作用及机制

摘要:
目的:观察蜂针疗法对佐剂性关节炎(adjuvant induced arthritis,AIA)模型大鼠症状及肠道菌群分布的影响,探讨其作用机制。方法:将29只雄性SD大鼠随机分为正常组4只,正常蜂针组、模型组、蜂针组、揿针组、甲氨蝶呤组,每组各5只。正常蜂针组、蜂针组和揿针组选取足三里、天枢穴干预,甲氨蝶呤组用甲氨蝶呤灌胃给药,治疗30 d后,腹主动脉取血进行血液分析,收集后足进行CT成像,采集盲肠粪便进行16S rRNA测序,检测各组大鼠肠道菌群的物种组成及多样性。结果:与模型组相比,蜂针组大鼠体重增长快,足趾肿胀减轻,炎症细胞比例降低,造血功能改善,血小板数恢复正常,肝肾功能改善,血脂水平升高,骨质破坏缓解;肠道菌群物种丰富度增加,丰度得到调节,普雷沃氏菌、副拟杆菌、棒状杆菌的丰度降低,阿德勒克洛伊茨菌、瘤胃球菌、嗜冷菌、脱硫弧菌丰度增加。蜂针组中罗氏菌、拟杆菌属、链球菌属、嗜血杆菌属、韦荣氏球菌属较其他各组丰度高。结论:蜂针疗法对AIA模型大鼠有良好的干预效果,且可以调节大鼠肠道菌群的分布,调节物种丰度,改善肠道环境。

Interventional effect of bee venom acupuncture on rats with adjuvant-induced arthritis and its mechanism:A study based on intestinal flora

Abstract:
Objective:To investigate the effect and mechanism of action of bee venom acupuncture on symptoms and intestinal flora distribution in rats with adjuvant-induced arthritis (AIA).Methods:A total of 29 male Sprague-Dawley (SD) rats were randomly divided into normal group with 4 rats,normal bee venom acupuncture group with 5 rats,model group with 5 rats,bee venom acupuncture group with 5 rats,press needle group with 5 rats,and methotrexate group with 5 rats.The rats in the normal bee venom acupuncture group,the bee venom acupuncture group,and the press needle group were given intervention at Zusanli and Tianshu points,and those in the methotrexate group were given methotrexate by gavage.After 30 days of treatment,blood samples were collected from the abdominal aorta to perform blood analysis;CT imaging was performed for hind foot;cecal fecal samples were collected,and 16S rRNA sequencing was performed to analyze the species composition and diversity of intestinal flora.Results:Compared with the model group,the bee venom acupuncture group had a rapid increase in body weight,alleviation of toe swelling,a reduction in the proportion of inflammatory cells,a platelet count returning to normal,improvements in hematopoietic function and hepatic and renal function,increases in blood lipid levels,and alleviation of bone destruction;the bee venom acupuncture group had significant increases in the abundances of intestinal flora species,with reductions in the abundances of Prevotella,Parabacteroides,and Corynebacterium and increases in the abundances of Adlercreutzia,Ruminococcus,Psychrobacter,and Desulfovibrio.The bee venom acupuncture group had significantly higher abundances of Roseburia,Bacteroides,Streptococcus,Haemophilus,and Veillonella than the other groups.Conclusion:Bee venom acupuncture has a marked therapeutic effect on rats with AIA and can regulate the distribution of intestinal flora,modulate the abundance of species,and improve intestinal environment.
