雷火灸对脑缺血大鼠神经功能及缺血半暗带NGF、TrkA表达的影响

摘要:
目的:观察雷火灸对脑缺血大鼠神经功能、相对脑缺血体积、缺血半暗带形态和神经生长因子(nerve growth factor,NGF)、原肌球蛋白受体激酶A(tropomyosin receptor kinase A,TrkA)表达的影响。方法:按随机数字表法将30只SD大鼠分为假手术组、模型组、雷火灸组,每组各10只;模型组、雷火灸组采用50%三氯化铁(ferric chloride,FeCl3)化学制备大脑中动脉血栓模型,雷火灸组给予艾灸“百会”干预,30 min/次,1次/d,连续干预7 d。采用Longa评分法评估各组大鼠神经功能,2,3,5-氯化三苯基四氮唑(2,3,5-triphenyltetrazolium chloride,TTC)染色法检测相对脑缺血体积,苏木精-伊红(hematoxylin and eosin,HE)染色法观察缺血半暗带组织形态变化,免疫组织化学法检测缺血半暗带NGF、TrkA的表达。结果:与假手术组比较,模型组大鼠Longa评分升高(P<0.01),相对脑缺血体积增大(P<0.01),缺血半暗带细胞构型紊乱,神经元大量变性坏死,NGF、TrkA表达增加(P<0.05)。与模型组比较,雷火灸组大鼠Longa评分降低(P<0.05),相对脑缺血体积缩小(P<0.05),缺血半暗带神经元损伤程度减轻,NGF、TrkA表达增加(P<0.05)。结论:雷火灸可能通过提高缺血半暗带NGF、TrkA的表达水平,改善脑缺血大鼠神经功能,缩小脑缺血体积,减轻脑损伤。

Effect of thunder-fire moxibustion on neurological function and the expression of nerve growth factor and tropomyosin receptor kinase A in the ischemic penumbra in a rat model of cerebral ischemia

Abstract:
Objective:To investigate the effect of thunder-fire moxibustion on neurological function,relative cerebral ischemic volume,ischemic penumbra morphology,and the expression of nerve growth factor (NGF) and tropomyosin receptor kinase A (TrkA) in a rat model of cerebral ischemia.Methods:A total of 30 Sprague-Dawley rats were divided into sham-operation group,model group,and thunder-fire moxibustion group using a random number table,with 10 rats in each group.The rats in the model group and the thunder-fire moxibustion group were treated with 50% ferric chloride to establish a model of middle cerebral artery thrombosis,and those in the thunder-fire moxibustion group were given moxibustion at Baihui point for 30 minutes each time,once a day for 7 consecutive days.The Longa scoring method was used to assess neurological function;2,3,5-triphenyltetrazolium chloride staining was used to measure relative cerebral ischemic volume,and HE staining was used to observe histomorphological changes of ischemic penumbra;immunohistochemistry was used to measure the expression levels of NGF and TrkA in ischemic penumbra.Results:Compared with the sham-operation group,the model group had significant increases in Longa score (P < 0.01),relative cerebral ischemic volume (P < 0.01),and the expression levels of NGF and TrkA in ischemic penumbra (P < 0.05),with disordered cell structure and massive degeneration and necrosis of neurons.Compared with the model group,the thunder-fire moxibustion group had significant reductions in Longa score (P < 0.05) and relative cerebral ischemic volume (P < 0.05),as well as alleviation of neuronal injury and increases in the expression levels of NGF and TrkA in ischemic penumbra (P < 0.05).Conclusion:Thunder-fire moxibustion can improve neurological function,reduce cerebral ischemic volume,and alleviate brain injury in rats with cerebral ischemia by increasing the expression levels of NGF and TrkA in ischemic penumbra.
