目的:基于数据挖掘筛选中医药防治冠心病室性早搏的核心药物,利用网络药理学探讨其作用机制。方法:检索中国知网(CNKI)、万方数据(WANFANG DATA)、维普中文科技期刊数据库(VIP)中有关中医药防治冠心病室性早搏的文献,使用Excel表格和古今医案云平台V2.3.5进行频次统计、关联、聚类和网络分析。利用TCMSP数据库获取核心药物主要化学成分,结合Swiss Target Prediction数据库预测活性成分作用靶点;运用OMIM、GeneCards、DrugBank等数据库获取冠心病室性早搏相关靶点;采用UniProt数据库匹配相关靶点基因;运用Cytoscape 3.9.1软件构建蛋白质-蛋白质相互作用网络图;采用Metascape平台进行基因本体(GO)功能、京都基因与基因组百科全书(KEGG)通路富集分析。结果:共筛选出124篇文献,包含124首方,涉及160味中药。高频药物排前6位的依次为甘草、丹参、酸枣仁、当归、黄芪、党参。药味以甘、苦、辛为主;药性以温、平、微寒、寒为主;归经以心、肝、脾、肺为主。获得18组关联药物、4首聚类处方。获得核心药物有效成分78种、药物靶点326个、疾病靶点1609个、疾病-药物交集靶点141个。GO功能富集分析获得生物过程5526个、细胞组分474个、分子功能787个,KEGG通路富集分析获得253条信号通路。结论:防治冠心病室性早搏的中药主要通过调控白细胞介素-6、肿瘤坏死因子、蛋白激酶Bα等核心关键靶点,糖尿病并发病中晚期糖基化产物-晚期糖基化终末产物受体信号通路、松弛素信号通路及钙信号通路等来发挥治疗冠心病室性早搏的作用。 |

Abstract: |
Objective:To investigate the core traditional Chinese medicine (TCM) drugs in the prevention and treatment of ventricular premature beats in coronary heart disease based on data mining,as well as their mechanism of action based on network pharmacology.Methods:CNKI,WANFANG DATA,and VIP were searched for articles on TCM in the prevention and treatment of ventricular premature beats in coronary heart disease,and Excel and Cloud Platform of Ancient and Modern Medical Records (V2.3.5) were used for frequency statistics,association analysis,cluster analysis,and network analysis.TCMSP database was used to obtain the main chemical components of core drugs,and Swiss Target Prediction database was used to predict the action targets of active components;OMIM,GeneCards,and DrugBank databases were used to obtain the targets associated with ventricular premature beats in coronary heart disease;UniProt database was used to obtain related target genes;Cytoscape 3.9.1 was used to construct a protein-protein interaction network;Metascape platform was used to perform gene ontology (GO) functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis.Results:A total of 124 articles were screened out,involving 124 prescriptions and 160 TCM drugs.The top 6 high-frequency drugs were Radix Glycyrrhizae,Salvia miltiorrhiza,spina date seed,Angelica sinensis,Astragalus membranaceus and Codonopsis pilosula.Most drugs had a sweet,bitter or pungent taste and were warm,neutral,slightly cold or cold in nature,and they mainly entered the heart,liver,spleen,and lung meridians.A total of 18 groups of associated drugs and 4 clustered prescriptions were obtained,and related analyses obtained 78 effective constituents of core drugs,326 drug targets,1609 disease targets,and 141 disease-drug intersecting targets.GO functional enrichment analysis obtained 5526 biological processes,474 cellular components,and 787 molecular functions,and KEGG pathway enrichment analysis obtained 253 signaling pathways.Conclusion:TCM drugs for the treatment and treatment of ventricular premature beats in coronary heart disease exert a therapeutic effect on ventricular premature beats possibly by regulating the key targets such as interleukin-6,tumor necrosis factor,and protein kinase-Bα through the signaling pathways including the AGE-RAGE signaling pathway in diabetic complications,the relaxin signaling pathway,and the calcium signaling pathway. |
