基于超声波提取的湘玉竹多糖工艺研究

摘要:
目的:优化湘玉竹多糖的提取工艺,提高玉竹多糖提取率。方法:采用超声辅助提取方法,通过 设定不同超声温度、超声时间和超声功率以优化湘玉竹多糖的提取工艺,设计单因素试验与正交试验相结合 的方法,系统探究超声波辅助提取湘玉竹多糖的关键工艺参数。结果:超声波辅助提取玉竹多糖最优条件组 合为:料液比1 ∶ 20(g/mL)、超声波处理温度45 ℃、作用时间10 min、功率强度350 W,YZ6 的提取率可达 12.32%。采用传统热水提取法对YZ6 进行提取,玉竹多糖的提取率为5.38%。在最佳提取工艺条件下对6 份 湖南不同地理来源的玉竹进行多糖含量测定,结果发现YZ3 多糖提取率最高,为26.00% ;YZ6 多糖提取率最 低,为12.32%。结论:与传统热水提取法相比,采用超声波辅助提取法可提高玉竹多糖提取率,提取最佳条件 为温度45℃、作用时间10 min、功率强度350 W。在此条件下检测发现来YZ3 多糖提取率最高,YZ6 多糖提 取率最低。

Extraction process of polysaccharides from Polygonatum odoratum produced in Hunan, China basedon ultrasonic extraction

Abstract:
Objective: To optimize the extraction process of polysaccharides from Polygonatum odoratum produced in Hunan, China, and to improve the extraction rate of Polygonatum odoratum polysaccharides. Methods: With the method of ultrasonic-assisted extraction, different ultrasonic temperatures, ultrasonic durations, and ultrasonic powers were set to optimize the extraction process of Polygonatum odoratum polysaccharides, and single-factor experiments and orthogonal experiments were used to determine the key process parameters for ultrasonic-assisted extraction of Polygonatum odoratum polysaccharides. Results: The optimal conditions for ultrasonic-assisted extraction of Polygonatum odoratum polysaccharides were a material-to-liquid ratio of 1∶20 (g/mL), an ultrasonic treatment temperature of 45℃ a duration of treatment of 10 minutes, and a power intensity of 350 W, and the extraction rate of YZ6 reached 12.32% under these conditions, while traditional hot water extraction yielded an extraction rate of 5.38% for Polygonatum odoratum polysaccharides. Content determination was performed for polysaccharides in six Polygonatum odoratum samples from different geographical sources in Hunan under the optimal extraction conditions, and the results showed that polysaccharide in YZ3 had the highest extraction rate of 26.00%, while YZ6 polysaccharide had the lowest extraction rate of 12.32%. Conclusion: Compared with the traditional hot water extraction method, ultrasonic-assisted extraction can improve the extraction rate of Polygonatum odoratum polysaccharides. The optimal extraction conditions are a temperature of 45℃, a duration of treatment of 10 minutes, and a power intensity of 350 W. Polysaccharide in YZ3 has the highest extraction rate and that in YZ6 has the lowest content under these conditions.
