基于网络药理学探讨扫日劳胶囊治疗急性肺损伤的作用机制

摘要:
目的:通过实验研究探索扫日劳胶囊干预急性肺损伤(acute lung injury,ALI)的疗效,网络药理学探讨其作用机制。方法:通过动物实验观察扫日劳胶囊对ALI小鼠肺脏指数、肺脏干/湿比重、肺脏组织形态和纤维化程度、肺脏组织内氧化性损伤相关的活性氧(reactive oxygen species,ROS)和丙二醛(malondialdehyde,MDA)含量及抗氧化相关酶总抗氧化能力(total antioxidant capacity,T-AOC)、超氧化物歧化酶(superoxide dismutase,SOD)、过氧化氢酶(catalase,CAT)含量的影响;通过中药系统药理学数据库与分析平台(Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform,TCMSP)、中药综合数据库(Traditional Chinese Medicine Integrated Database,TCMID)、疾病基因网络数据库(DisGeNET)、在线人类孟德尔遗传数据库(Online Mendelian Inheritance in Man,OMIM)、基因卡片数据库(GeneCards),构建扫日劳-急性肺损伤核心靶点网络,通过基因本体(gene ontology,GO)功能和京都基因与基因组数据库(Kyoto Encyclopedia of Genes and Genomes,KEGG)通路富集分析确定扫日劳胶囊治疗ALI的重要靶点和信号通路。结果:与模型组比较,扫日劳中、高剂量组小鼠肺脏指数下降(P<0.01),肺脏干/湿比重增加(P<0.01),肺组织病理损伤程度和纤维化程度减轻,组织内ROS和MDA含量显著下降(P<0.01),T-AOC、SOD、CAT水平增加(P<0.01)。筛选得到ALI与扫日劳共同靶点221个、核心靶点16个,GO功能富集分析获得2 037个条目,KEGG通路富集分析获得181条通路。结论:扫日劳胶囊具有治疗急性肺损伤的作用,其作用的发挥与磷脂酰肌醇3-激酶-蛋白激酶B(Phosphatidylinositol 3-kinase-Protein Kinase B,PI3K-AKT)信号通路密切相关。

Abstract:
Objective:To investigate the effect of Saorilao capsules in the treatment of acute lung injury (ALI) and its mechanism of action based on network pharmacology.Methods:Animal experiments were conducted to observe the effect of Saorilao capsules on lung index,lung dry/wet weight ratio,lung histomorphology,lung fibrosis degree,the content of reactive oxygen species (ROS) and malondialdehyde (MDA) associated with oxidative damage in lung tissue,the total antioxidant capacity (T-AOC) of antioxidant enzymes,and the content of superoxide dismutase (SOD) and catalase (CAT).Based on the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform,Traditional Chinese Medicine Integrated Database,DisGeNET,Online Mendelian Inheritance in Man,and GeneCards database,a Saorilao-ALI core target network was constructed,and the gene ontology (GO) functional enrichment analysis and the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis were performed to identify the important targets and signaling pathways of Saorilao capsules in the treatment of ALI.Results:Compared with the model group,the middle- and high-dose Saorilao groups had a significant reduction in lung index (P < 0.01),a significant increase in lung dry/wet weight ratio (P < 0.01),alleviation of lung histopathological injury and fibrosis degree,significant reductions in the content of ROS and MDA (P < 0.01),and significant increases in the levels of T-AOC,SOD,and CAT (P < 0.01).A total of 221 common targets were obtained for ALI and Saorilao,with 16 core targets.The GO functional enrichment analysis obtained 2037 terms,and the KEGG pathway enrichment analysis obtained 181 pathways.Conclusion:Saorilao capsules exert a therapeutic effect on ALI through the phosphatidylinositol 3-kinase-protein kinase B signaling pathway.
