目的:探讨复方藜芍片(Fufang Lishao tablets,FLS)对原发性三叉神经痛(primary trigeminal neuralgia,PTN)大鼠疼痛改善作用和对脑源性神经营养因子(brain-derived neurotrophic factor,BDNF)及其受体酪氨酸激酶受体B(tyrosine kinase receptor B,TrkB)表达的影响。方法:采用慢性压迫性损伤眶下神经术对60只雄性SD大鼠建立PTN模型,随机分为模型组、卡马西平组(20 mg/kg)、复方藜芍片低剂量组(FLS-L组,420 mg/kg)、复方藜芍片中剂量组(FLS-M组,840 mg/kg)、复方藜芍片高剂量组(FLS-H组,1 680 mg/kg),每组12只。对各组大鼠进行动物行为学评估,测定大鼠术侧面部机械痛敏阈值(mechanical withdrawal threshold,MWT); ELISA法测定血清中白细胞介素- 6(interleukin-6,IL-6)、BDNF水平。Western blot测定三叉神经节(trigeminal ganglion,TG)中BDNF、TrkB表达水平。结果:与术前相比,手术后当天各组大鼠MWT值降低(P<0.05),术后第28 d模型组MWT值为8.25,大鼠PTN模型造模成功;自术后第11天起,FLS-M、FLS-H组大鼠MWT值均高于与模型组(P<0.05);术后17~21 d,FLS-M、FLS-H组大鼠MWT值均高于卡马西平组(P<0.05)。与模型组相比,FLS-M、FLS-H组大鼠血清IL-6水平下降(P<0.05),复方藜芍片各剂量组大鼠血清BDNF及TG中BDNF、TrkB的表达水平均降低,差异均具有统计学意义(P<0.05)。结论:复方藜芍片能够改善原发性三叉神经痛大鼠的痛觉过敏现象,可能与减轻炎症反应和抑制BDNF及其受体TrkB的表达有关。 |

Effect of Fufang Lishao tablets on the expression of brain-derived neurotrophic factor and tyrosine kinase receptor B in rats with primary trigeminal neuralgia |

Objective:To investigate the effect of Fufang Lishao tablets (FLS) in improving pain in rats with primary trigeminal neuralgia (PTN),as well as its effect on the expression of brain-derived neurotrophic factor (BDNF) and its receptor tyrosine kinase receptor B (TrkB).Methods:A total of 60 male Sprague-Dawley rats were selected,and the method of chronic constriction injury of the infraorbital nerve was used to establish a model of PTN.The rats were randomly divided into model group,carbamazepine (20 mg/kg) group,low-dose FLS group (FLS-L group,420 mg/kg),middle-dose FLS group (FLS-M group,840 mg/kg),and high-dose FLS group (FLS-H group,1 680 mg/kg),with 12 rats in each group.Behavioral assessment was performed for the rats in each group,and mechanical withdrawal threshold (MWT) of face at the surgical side was measured;ELISA was used to measure the serum levels of interleukin-6 (IL-6) and BDNF;Western blot was used to measure the expression level of BDNF and TrkB in the trigeminal ganglion (TG).Results:After surgery,all groups had a significant reduction in MWT on the day of surgery (P<0.05),with an MWT value of 8.25 on day 28 after surgery,suggesting that the rat model of PTN was successfully established.Since day 11 after surgery,the FLS-M group and the FLS-H group had a significantly higher MWT value than the model group (P<0.05),and on days 17-21 after surgery,the FLS-M group and the FLS-H group had a significantly higher MWT value than the carbamazepine group (P<0.05).Compared with the model group,the FLS-M group and the FLS-H group had a significant reduction in the serum level of IL-6 (P<0.05),and each FLS dose group had significant reductions in the serum level of BDNF and the expression levels of BDNF and TrkB in the TG (P<0.05).Conclusion:FLS can improve hyperalgesia in rats with PTN,possibly by alleviating inflammatory response and inhibiting the expression of BDNF and its receptor TrkB. |
