基于网络药理学探讨散结消瘤方治疗乳腺癌的作用机制

摘要:
目的:基于网络药理学方法探究散结消瘤方治疗乳腺癌的主要作用靶标和主要通路。方法:通过TCMSP平台检索散结消瘤方的活性成分及其对应靶标。利用UniProt和GeneCards数据库获得潜在靶标基因。使用Cytoscape 3.9.0软件构建散结消瘤方的“成分-靶标”网络。使用Bioinformatics平台将药物靶蛋白和疾病靶点相映射,并绘制Venny图。结合STRING数据库及Cytoscape软件构建蛋白质-蛋白质相互作用网络。利用Database for Annotation、DAVID生物信息资源数据库对散结消瘤方的作用靶标进行基因本体(GO)功能和京都基因与基因组百科全书(KEGG)通路富集分析。结果:获得散结消瘤方主要活性成分196种,包括β-谷甾醇、木犀草素、甘草查尔酮及槲皮素等,其中与乳腺癌相关的作用靶标有112个,包括肿瘤蛋白P53、雌激素受体1、丝氨酸特异性蛋白激酶1、原癌基因、氨基端激酶等。涉及的生物过程为激素应答、无机物反应等;细胞组分为激酶结合、蛋白质激酶结合等;分子功能为转录调节复合物、膜筏等。主要涉及癌症途径、脂质和动脉粥样硬化、磷脂酰肌醇-3-激酶/蛋白激酶信号、乙型肝炎等通路。结论:散结消瘤方治疗乳腺癌具有多靶点、多机制的优势,为散结消瘤方治疗乳腺癌具体作用机制和新药的研发提供依据。

Mechanism of action of Sanjie Xiaoliu prescription in treatment of breast cancer:A study based on network pharmacology

Abstract:
Objective:To investigate the main action targets and pathways of Sanjie Xiaoliu prescription in the treatment of breast cancer based on network pharmacology.Methods:TCMSP platform was used to search for the active components of Sanjie Xiaoliu prescription and their corresponding targets.UniProt and GeneCards databases were used to obtain potential target genes.Cytoscape 3.9.0 software was used to construct a component-target network for Sanjie Xiaoliu prescription.Bioinformatics platform was used for the mapping of drug target proteins and disease targets,and Venny diagram was plotted.STRING database and Cytoscape software were used to construct a protein-protein interaction network.Database for Annotation and DAVID bioinformatic resource database were used to perform gene ontology (GO) functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis of the action targets of Sanjie Xiaoliu prescription.Results:A total of 196 main active components were obtained for Sanjie Xiaoliu prescription,including beta-sitosterol,luteolin,licochalcone,and quercetin,among which there were 112 action targets associated with breast cancer,including tumor protein P53,estrogen receptor 1,serine-specific protein kinase 1,protooncogene,and JNK.The biological processes involved included hormone response and inorganic reaction;cellular components included kinase binding and protein kinase binding;molecular functions included transcriptional regulation complex and membrane raft.The pathways involved included the cancer pathway,lipids and atherosclerosis,the phosphatidylinositol 3-kinase/protein kinase signaling pathway,and the hepatitis B pathway.Conclusion:Sanjie Xiaoliu prescription has the advantages of multiple targets and mechanisms in the treatment of breast cancer,which provides a basis for the research on the specific mechanism of action of Sanjie Xiaoliu prescription in the treatment of breast cancer and the research and development of related new drugs.
