茶多酚对口臭患者唾液细菌多样性的影响

摘要:
目的:评价茶多酚在口臭患者口腔微生态中的作用,通过分析样本内细菌群落结构差异讨论茶多酚对于口臭的疗效。方法:收集6位口臭患者的唾液,混合均匀后取4份5ml的样本,分别与阳性组、实验组、阴性组和空白组的TPY培养基混合后培养48h。最后进行16S rDNA V3-V4区基因组测序分析。结果:阳性组和实验组均能促进样本细菌OTU数的恢复。在门水平上,阳性组Firmicutes的相对丰度显著低于其余3组(P<0.01),而Actinobacteria的相对丰度显著高于其余3组(P<0.01);实验组Firmicutes的相对丰度显著低于空白组(P<0.01),Actinobacteria的相对丰度明显低于阴性组(P<0.05)。阳性组和实验组Proterbacteria的相对丰度显著高于阴性组和空白组(P<0.01),阳性组Bacteroidetes的相对丰度明显高于空白组(P<0.05)。从细菌目分类比较,各组Lactobacillales的相对丰度显著不同(P<0.01),从高到低依次为实验组、阳性组、阴性组、空白组。实验组Bacillales的相对丰度显著低于空白组(P<0.05);阳性组Actinomycetales的相对丰度显著高于空白组和实验组(P<0.05);实验组Pesudomonadales的相对丰度显著高于空白组(P<0.05)。结论:茶多酚通过对细菌门、目分类组成比例的影响来达到治疗口臭的效果,其疗效不及西吡氯铵含片。

Effect of tea polyphenol on bacterial diversity in saliva of patients with fetid breath

Abstract:
Objective:To investigate the role of tea polyphenol in oral microecology in patients with fetid breath and the clinical effect of tea polyphenol in the treatment of fetid breath based on an analysis of bacterial community structure in samples.Methods:Saliva was collected from 6 patients with fetid breath and was mixed up thoroughly,and then 4 samples with a volume of 5 mL each were prepared.The 4 samples were mixed with the TPY medium in positive group,experimental group,negative group,and blank group,respectively,and were cultured for 48 hours.Genome sequencing was performed for the V3 and V4 regions of 16S rDNA.Results:Recovery of the operational taxonomic unit number of bacteria was promoted in the positive group and the experimental group.At the phylum level,the positive group had a significantly lower relative abundance of Firmicutes and a significantly higher relative abundance of Actinobacteria than the other three groups (P<0.01),and the experimental group had a significantly lower relative abundance of Firmicutes than the blank group (P<0.01) and a significantly lower relative abundance of Actinobacteria than the negative group (P<0.05).The positive group and the experimental group had a significantly higher relative abundance of Proteobacteria than the negative group and the blank group (P<0.01),and the positive group had a significantly higher relative abundance of Bacteroidetes than the blank group (P<0.05).At the order level,there was a significant difference in the relative abundance of Lactobacillales between groups (P<0.01),and the experimental group had the highest relative abundance,followed by the positive group,the negative group,and the blank group.The experimental group had a significantly lower relative abundance of Bacillales than the blank group (P<0.05);the positive group had a significantly higher relative abundance of Actinomycetales than the blank group and the experimental group (P<0.05);the experimental group had a significantly higher relative abundance of Pseudomonadales than the blank group (P<0.05).Conclusion:Tea polyphenol exerts a therapeutic effect on fetid breath by affecting the composition of bacteria at the phylum and order levels,with a lower clinical effect than cetylpyridinium chloride buccal tablets.
