肺痨康对COPD模型大鼠脏器系数的影响

摘要:
目的:观察肺痨康对慢性阻塞性肺疾病(chronic obstructive pulmonary disease,COPD)模型大鼠脏器系数的影响,探究其对COPD模型大鼠是否具有治疗作用及其作用机制。方法:将60只SD雄性大鼠适应性喂养1周后随机分为空白组、模型组、地米塞松组及肺痨康高、中、低剂量组,采用香烟烟熏(CS)+脂多糖(LPS)气管内滴注复合因素法建立COPD大鼠模型,于造模第15天开始,肺痨康低、中、高剂量组分别予肺痨康药液0.1405、0.281、0.843ml/(kg·d)灌胃,地塞米松组予地塞米松每天每只0.054mg灌胃,空白组、模型组予相应体积0.9%氯化钠注射液灌胃,每天1次。灌胃14d后处死、取材、称重并计算相关脏器系数。结果:与模型组比较,地塞米松组、肺痨康高剂量组能不同程度地降低肺脏系数(P<0.01),提高脾脏系数和胸腺系数(P<0.01,P<0.05),肺痨康中、低剂量组能提高胸腺系数(P<0.01)。结论:肺痨康高剂量可减轻模型大鼠肺脏的炎症反应并增强其免疫力。

Effect of Feilaokang on organ coefficient in rats with chronic obstructive pulmonary disease

Abstract:
Objective:To investigate the effect of Feilaokang on organ coefficient in rats with chronic obstructive pulmonary disease(COPD),as well as its therapeutic effect on COPD and possible mechanism of action.Methods:After one week of adaptive feeding,60 Sprague-Dawley rats were randomly divided into blank group,model group,dexamethasone group,and high-,middle-,and low-dose Feilaokang groups.A rat model of COPD was established by cigarette smoke combined with intratracheal instillation of lipopolysaccharide.From day 15 of modeling,the rats in the low-,middle-,and high-dose Feilaokang groups were given Feilaokang drug solution once a day by gavage at a dose of 0.1405,0.281,and 0.843 ml/kg/d,respectively,each rat in the dexamethasone group was given dexamethasone once a day by gavage at a dose of 0.054mg/d,and those in the blank group and the model group were given an equal volume of 0.9% sodium chloride injection once a day by gavage.The rats were sacrificed after 14 days of treatment by gavage,and related samples were collected and weighed to calculate the organ coefficient.Results:Compared with the model group,the dexamethasone group and the high-dose Feilaokang group had a significant reduction in lung coefficient(P<0.01)and significant increases in spleen coefficient and thymus index(P<0.01 or P<0.05),and the middle-and low-dose Feilaokang groups had a significant increase in thymus index(P<0.01).Conclusion:High-dose Feilaokang can alleviate lung inflammatory response and enhance immunity in rats with COPD.
