心痛方对急性心肌梗死大鼠NCX mRNA表达及Ca2+浓度的影响

摘要:
目的:探究心痛方对心肌梗死大鼠心肌组织内钠钙交换蛋白(NCX mRNA)的表达及钙离子(Ca2+)浓度的影响。方法:将50只SD大鼠分为假手术组、模型组、心痛方高剂量组、心痛方低剂量组、硫氮卓酮组,采用结扎大鼠冠状动脉左前降支的方法建立急性心肌梗死大鼠模型。运用RT-PCR法检测各组大鼠心肌细胞内NCX mRNA的表达,激光扫描共聚显微镜法检测大鼠心肌组织中的Ca2+浓度,HE染色法测定大鼠心肌梗死面积。结果:在下调NCX mRNA表达方面,心痛方高、低剂量组与假手术组、模型组、硫氮卓酮组比较,差异有统计学意义(P<0.01);在降低MFI方面,心痛方高、低剂量组与假手术组、模型组比较,差异有统计学意义(P<0.05),且心痛方高剂量组在降低MFI方面还优于硫氮卓酮组(P<0.01);在减少心肌梗死面积方面,心痛方高、低剂量组与模型组、硫氮卓酮组比较,差异有统计学意义(P<0.05)。结论:心痛方能下调急性心肌梗死大鼠NCX mRNA的表达,降低胞内Ca2+的浓度,从而抑制钙超载对心肌细胞的损害,减少心肌梗死面积。

Abstract:
Objective:To investigate the effect of Xintong prescription on NCX mRNA expression and calcium ion(Ca22+)concentration in myocardial tissue in rats with acute myocardial infarction.Methods:A total of 50 Sprague-Dawley rats were divided into sham-operation group,model group,high-and low-dose Xintong prescription groups,and diltiazem group,and ligation of the left anterior descending coronary artery was performed to establish a rat model of acute myocardial infarction.RT-PCR was used to measure the mRNA expression of NCX in cardiomyocytes,laser scanning confocal microscopy was used to measure the concentration of Ca22+ in myocardial tissue,and HE staining was used to measure myocardial infarct area.Results:The high-and low-dose Xintong prescription groups had a significantly greater reduction in the mRNA expression of NCX than the sham-operation group,the model group,and the diltiazem group(P<0.01).The high-and low-dose Xintong prescription groups had a significantly greater reduction in mean fluorescence intensity than the sham-operation group and the model group(P<0.05),and the high-dose Xintong prescription group had a significantly greater reduction than the diltiazem group(P<0.01).The high-and low-dose Xintong prescription groups had a significantly greater reduction in myocardial infarct area than the model group and the diltiazem group(P<0.05).Conclusion:Xintong prescription can downregulate the mRNA expression of NCX,reduce the concentration of Ca22+,inhibit cardiomyocyte damage due to calcium overload,and thus reduce myocardial infarct area.
