| 目的:研究健脾补肾方对结直肠癌裸鼠中消皮素D(gasdermin D,GSDMD)、半胱天冬酶-1(cysteine-dependent aspartate-specific protease 1,Caspase-1)和白细胞介素-1β(interleukin-1β,IL-1β)蛋白表达和肿瘤细胞焦亡的影响。方法:构建人结直肠癌裸鼠移植瘤模型,将其分为中药组和对照组。中药组予健脾补肾方灌胃干预,对照组予0.9%氯化钠注射液灌胃干预,2组均隔天给药1次,连续干预14 d。记录裸鼠体重、移植瘤的质量和体积,酶联免疫吸附试验(Enzyme-Linked Immunosorbent Assay,ELISA)检测裸鼠血清中IL-1β和白细胞介素-18(interleukin-18,IL-18)的含量,免疫组化法检测肿瘤GSDMD、Caspase-1和IL-1β的表达。结果:与对照组相比,中药组裸鼠的体重、移植瘤的质量和体积均降低(P<0.01),血清IL-1β、IL-18含量增加(P<0.01),肿瘤细胞GSDMD、Caspase-1、IL-1β的表达增加(P<0.01)。结论:健脾补肾方可抑制结直肠癌裸鼠移植瘤生长,并伴随GSDMD、Caspase-1和IL-1β表达升高及血清炎症因子水平增加,提示其抗肿瘤作用可能与促进细胞焦亡相关。 |

| Effect of Jianpi Bushen prescription on the expression of gasdermin D,cysteine-dependent aspartate-specific protease 1,and interleukin-1β in nude mouse xenograft tumor of human colorectal cancer |

| Objective:To investigate the effect of Jianpi Bushen prescription on the protein expression of gasdermin D (GSDMD),cysteine-dependent aspartate-specific protease 1 (Caspase-1),and interleukin-1β (IL-1β) and tumor cell pyroptosis in nude mice with colorectal cancer.Methods: The nude mouse xenograft tumor model of human colorectal cancer was established,and the mice were divided into traditional Chinese medicine (TCM) group and control group.The mice in the TCM group were given Jianpi Bushen prescription by gavage,and those in the control group were given 0.9% sodium chloride injection by gavage,once every other day for 14 consecutive days.The body weight of nude mice was recorded,as well as the weight and volume of xenograft tumor;ELISA was used to measure the serum levels of IL-1β and interleukin-18 (IL-18),and immunohistochemistry was used to measure the expression of GSDMD,Caspase-1,and IL-1β in tumor.Results: Compared with the control group,the TCM group had significant reductions in the body weight of nude mice and weight and volume of xenograft tumor (P < 0.01) and significant increases in the serum levels of IL-1β and IL-18 (P < 0.01) and the expression levels of GSDMD,Caspase-1,and IL-1β in tumor cells (P < 0.01).Conclusion: Jianpi Bushen prescription can inhibit the growth of nude mouse xenograft tumor of colorectal cancer,with increases in the expression levels of GSDMD,Caspase-1,and IL-1β and the serum levels of inflammatory factors,suggesting that Jianpi Bushen prescription might exert an antitumor effect by promoting cell pyroptosis. |
