目的:以Wnt/β-连环蛋白(Wnt/β-catenin)信号轴为研究对象,探讨黄芩泻白散对支气管哮喘(简称哮喘)小鼠的治疗作用及其机制。方法:将60只小鼠根据随机分组原则分为正常对照组、模型组、地塞米松组及黄芩泻白散低、中、高剂量组。除正常对照组以外的其他组小鼠均采用卵清蛋白(ovalbumin,OVA)致敏及激发法建立哮喘模型,地塞米松组及黄芩泻白散各剂量组分别给予相应药物进行干预。观察各组小鼠的精神状态、饮食与呼吸等行为学表现。HE染色后用光镜观察小鼠肺组织的病理形态;全自动血液分析仪对小鼠支气管肺泡灌洗液(bronchoalveolar lavage fluid,BALF)进行炎症细胞分类计数;酶联免疫吸附分析法(enzymelinked immunosorbent assay,ELISA)检测小鼠BALF中白细胞介素-4(interleukin-4,IL-4)、白细胞介素-13(interleukin-13,IL-13)和白细胞介素-5(interleukin-5,IL-5)的含量;反转录聚合酶链反应(reverse trans cription polymerase chain reaction,RT-qPCR)法检测小鼠肺组织β-连环蛋白、糖原合成酶激酶3βmRNA的表达。结果:与正常对照组比较,模型组精神状态及饮食情况明显变差,出现咳嗽、气促等呼吸道症状;肺组织病理形态异常;BALF中炎症细胞数量及IL-4、IL-13、IL-5含量升高(P<0.01);肺组织β-连环蛋白mRNA表达升高,糖原合成酶激酶3β mRNA表达降低(P<0.01)。与模型组比较,地塞米松组、黄芩泻白散各剂量组精神状态及饮食情况改善,咳嗽、气促等呼吸道症状缓解,肺组织损害程度减轻,BALF中的炎症细胞数量及IL-4、IL-13、IL-5含量下降(P<0.01);肺组织β-连环蛋白mRNA表达升高,糖原合成酶激酶3βmRNA表达降低(P<0.01)。结论:黄芩泻白散可有效改善哮喘小鼠症状,对哮喘小鼠有治疗作用,其作用可能与调节Wnt/β-catenin信号通路活性有关。 |

Therapeutic effect of Huangqin Xiebai powder on rats with bronchial asthma and its effect on the Wnt/β-catenin signaling axis |

Objective:To investigate the therapeutic effect and mechanism of Huangqin Xiebai powder on rats with bronchial asthma based on the Wnt/β-catenin signaling axis.Methods:A total of 60 mice were randomly divided into normal control group,model group,dexamethasone group,and low,middle-,and high-dose Huangqin Xiebai powder groups.All mice except those in the normal control group were used to establish a model of asthma by ovalbumin sensitization and challenge,and the mice in the dexamethasone group and the Huangqin Xiebai powder groups were given the corresponding drug for intervention.The mice in each group were assessed in terms of mental state and behavioral manifestations including food intake and respiration.After HE staining,a light microscope was used to observe the pathomorphology of lung tissue;an automatic blood analyzer was used for classification and counting of inflammatory cells in bronchoalveolar lavage fluid (BALF);ELISA was used to measure the levels of interleukin-4 (IL-4),interleukin-13 (IL-13),and interleukin-5 (IL-5)in BALF;RT-qPCR was used to measure the mRNA expression levels of β-catenin and glycogen synthase kinase-3β in lung tissue.Results:Compared with the normal control group,the model group had poor mental state and food intake,the respiratory symptoms of cough and shortness of breath,abnormal pathomorphology of lung tissue,and significant increases in the number of inflammatory cells and the levels of IL-4,IL-13,and IL-5 in BALF (all P<0.01),as well as a significant increase in the mRNA expression level of β-catenin and a significant reduction in the mRNA expression level of glycogen synthase kinase-3β in lung tissue (P<0.01).Compared with the model group,the dexamethasone group and Huangqin Xiebai powder groups had improvements in mental state,food uptake,the respiratory symptoms of cough and shortness of breath,and the degree of lung injury and significant reductions in the number of inflammatory cells and the levels of IL-4,IL-13,and IL-5 in BALF (P<0.01),as well as a significant increase in the mRNA expression level of β-catenin and a significant reduction in the mRNA expression level of glycogen synthase kinase-3β in lung tissue (P<0.01).Conclusion:Huangqin Xiebai powder can effectively improve symptoms and exert a therapeutic effect on rats with asthma,possibly by regulating the activity of the Wnt/β-catenin signaling pathway. |
