目的:观察马归液含药血清对膀胱癌T24细胞周期及P53/P21信号通路的影响,并探讨其可能的干预机制。方法:将膀胱癌T24细胞分为空白对照组、模型组(0.9%氯化钠注射液)、不同浓度马归液含药血清药物组(正常浓度、2倍浓度、4倍浓度、6倍浓度组),采用CCK-8法检测各组含药血清对细胞增殖的影响,流式细胞术检测马归液含药血清对膀胱癌T24细胞周期的阻滞作用,RT-qPCR技术测定马归液含药血清干预后的T24细胞中P53、P21蛋白的mRNA表达情况,蛋白免疫印迹法检测马归液含药血清干预后T24细胞中P53/P21蛋白表达情况。结果:马归液不同浓度药物组干预T24细胞72 h后细胞活性均下降,且下降程度与药物浓度呈正相关(P<0.05);马归液含药血清干预膀胱癌T24细胞72 h后,细胞周期G1期细胞比例升高,S+G2期细胞比例降低,升高与降低的程度与药物浓度呈正相关(P<0.05);马归液含药血清可不同程度上调P53 mRNA、P21 mRNA及蛋白的表达,且与浓度呈正相关(P<0.01)。结论:马归液可通过上调P53表达,进而上调P21的表达,抑制细胞周期蛋白依赖性激酶4(CDK4)发挥细胞周期调控的关键作用,将细胞周期进程阻于G1期,从而抑制膀胱癌T24细胞的异常增殖。 |

Objective:To investigate the effect and possible intervention mechanism of serum containing Magui liquid on cell cycle and the P53/P21 signaling pathway in bladder cancer T24 cells.Methods:Bladder cancer T24 cells were divided into blank control group,model group (0.9% sodium chloride injection),and serum containing Magui liquid groups (the normal concentration of Magui liquid and the 2-,4-,and 6-fold concentrations of Magui liquid).CCK-8 assay was used to observe the influence of drug-containing serum on cell proliferation in each group;flow cytometry was used to observe the cell cycle arrest effect of drug-containing serum in bladder cancer T24 cells;RT-qPCR was used to measure the mRNA expression levels of P53 and P21 in T24 cells after intervention with the serum containing Magui liquid,and Western blot was used to measure the protein expression levels of P53 and P21 in T24 cells after such intervention.Results:There was a reduction in the viability of T24 cells after 72 hours of intervention with different concentrations of Magui liquid,and the degree of reduction was positively correlated with drug concentration (P<0.05).After the intervention of bladder cancer T24 cells with serum containing Magui liquid for 72 hours,there was an increase in the proportion of cells arrested in the G1 phase and a reduction in the proportion of cells arrested in the S+G2 phase,and the degree of increase or reduction was positively correlated with drug concentration (P<0.05).The serum containing Magui liquid could upregulate the mRNA and protein expression levels of P53 and P21,which was positively correlated with drug concentration (P<0.01).Conclusion:Magui liquid plays a key role in cell cycle regulation by upregulating the expression of P53 and P21 and inhibiting cyclin-dependent kinase 4,and it can inhibit the abnormal proliferation of bladder cancer T24 cells by arresting cells in the G1 phase. |
