目的:基于网络药理学探讨“胡毓恒降压方”治疗高血压病的作用机制。方法:借助数据库获取“胡毓恒降压方”靶基因与高血压病发病基因的交集基因,利用Cytoscape 3.9.1软件构建中药化合物-靶点网络及蛋白质与蛋白质相互作用(PPI)网络筛选关键靶点,对关键靶点进行基因本体(GO)功能和京都基因与基因组百科全书(KEGG)通路富集分析。结果:筛选获得木犀草素、槲皮素、山柰酚和黄芩素等主要功效成分23个,获得“胡毓恒降压方”治疗高血压病的潜在靶点117个,PPI分析筛选得到类固醇受体共激活因子基因(SRC)、磷脂酰肌醇-3-激酶p85基因(PI3KR1)、蛋白酪氨酸磷酸酶非受体型11基因(PTPN11)、蛋白激酶B基因(AKT1)等59个核心基因。GO分析得出生物过程29条,涉及靶点较多的生物过程有G蛋白偶联受体信号通路、蛋白质磷酸化、蛋白激酶B信号的正向调节、正调节MAP激酶活性、跨膜受体蛋白酪氨酸激酶信号通路、炎症反应等。KEGG分析获得相关信号通路44条,涉及靶点较多的信号通路有神经活性配体-受体相互作用、钙信号通路、氮代谢、内分泌抵抗等。结论:“胡毓恒降压方”主要通过木犀草素、槲皮素、山柰酚和黄芩素等成分经多靶点、多途径、多通路发挥降血压的作用,该方治疗高血压病的机制可能与神经活性配体-受体相互作用、钙信号通路相关。该结论仍需通过细胞实验、动物实验等予以进一步验证。 |

Mechanism of action of Hu Yuheng’s antihypertensive prescription in treatment of hypertension:A study based on network pharmacology |

Abstract: |
Objective:To investigate the mechanism of action of Hu Yuheng’s antihypertensive prescription in the treatment of hypertension based on network pharmacology. Methods:Related databases were used to obtain the intersecting genes between the target genes of Hu Yuheng’s antihypertensive prescription and the genes of hypertension. Cytoscape 3.9.1 was used to construct a traditional Chinese medicine (TCM) compound-target network and a protein-protein interaction (PPI) network,and gene ontology (GO) functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis were performed for the key targets. Results:A total of 23 main active components were obtained,including luteolin,quercetin,kaempferol,and baicalein,and a total of 117 potential targets were obtained for Hu Yuheng’s antihypertensive prescription in the treatment of hypertension.The PPI analysis obtained 59 core genes,including SRC,PI3KR1,PTPN11,and AKT1.The GO analysis obtained 29 biological processes,among which the G protein-coupled receptor signaling pathway,protein phosphorylation,positive regulation of protein kinase-B signal,positive regulation of MAP kinase activity,the transmembrane receptor protein tyrosine kinase signaling pathway,and inflammatory response involved a relatively high number of targets.The KEGG analysis obtained 44 related signaling pathways,among which neuroactive ligand-receptor interaction,the calcium signaling pathway,nitrogen metabolism,and endocrine resistance involved a relatively high number of targets. Conclusion:Hu Yuheng’s antihypertensive prescription exerts an antihypertensive effect through multiple targets,paths,and pathways via the components such as luteolin,quercetin,kaempferol,and baicalein,and the mechanism of this prescription may be associated with neuroactive ligand-receptor interaction and the calcium signaling pathway. Further studies such as cell and animal experiments are needed to verify such results. |
