三七总皂苷对野百合碱诱导的PASMCs增殖、凋亡及ADAM10/Notch3信号通路的影响

摘要:
目的:探讨在野百合碱(MCT)作用下三七总皂苷(PNS)对大鼠肺动脉平滑肌细胞(PASMCs)增殖、凋亡及其解整合素金属蛋白酶10/神经源性基因座notch同源蛋白-3(ADAM10/Notch3)通路的影响。方法:将体外培养的PASMCs随机分为正常组(C组)、野百合碱组(M组)和野百合碱+三七总皂苷组(P组),采用CCK-8法测各组细胞活力;EdU法检测细胞增殖;qPCR法和Western blot法分别检测细胞ADAM10、Notch3、人源重组蛋白1(Hes-1)、细胞周期蛋白依赖性激酶抑制因子(p27)、增殖细胞核抗原(PCNA)、半胱氨酸天冬氨酸蛋白酶3(Caspase-3)mRNA和蛋白的表达情况。结果:在MCT作用下,PASMCs细胞活力增强,EdU阳性率增加;ADAM10、Notch3、Hes-1、PCNA的mRNA以及蛋白表达上调(P<0.05,P<0.01),p27和Caspase-3的mRNA以及蛋白表达下调(P<0.05,P<0.01);使用PNS可下调MCT作用下的上述作用(P<0.05,P<0.01)。结论:三七总皂苷可通过抑制ADAM10/Notch3信号通路,减弱大鼠PASMCs增殖、促进PASMCs凋亡。

Abstract:
Objective:To investigate the effect of Panax notoginseng saponins (PNS) on the proliferation and apoptosis of rat pulmonary artery smooth muscle cells and the ADAM10/Notch3 signaling pathway under the action of monocrotaline (MCT).Methods:PASMCs cultured in vitro were randomly divided into normal group (group C),MCT group (group M),and MCT+PNS group (group P),and CCK-8 assay was used to measure cell viability;the EdU method was used to measure cell proliferation;qPCR and Western blot were used to measure the mRNA and protein expression levels of ADAM10,Notch3,Hes-1,p27,proliferating cell nuclear antigen (PCNA),and caspase-3.Results:Under the action of MCT,there were increases in the viability of PASMCs,EdU positive rate,and the mRNA and protein expression levels of ADAM10,Notch3,Hes-1,and PCNA(P<0.05,P<0.01),and there were reductions in the mRNA and protein expression levels of p27 and caspase-3(P<0.05,P<0.01),while PNS could reduce the above effects under the action of MCT(P<0.05,P<0.01).Conclusion:PNS can weaken the proliferation of rat PASMCs and promote their apoptosis by inhibiting the ADAM10/Notch3 signaling pathway.
