目的:利用GEO数据库和网络药理学分析猫爪草治疗原发性肝癌(HCC)的作用机制。方法:借助HERB数据库、SwissADME及SwissTargetPrediction筛选猫爪草的有效成分与作用靶点。从GEO数据库中筛选正常肝脏组织患者与肝癌患者的差异表达基因,整合GeneCards、DisGeNET、GEO数据库得到HCC疾病靶点、疾病和成分的交集基因,利用Cytoscape 3.9.0构建成分-疾病网络及拓扑分析,在David数据库进行基因本体(GO)功能、京都基因与基因组百科全书(KEGG)通路富集分析。结果:获得猫爪草活性成分36种,潜在靶点544个,差异表达基因1 950个。核心靶点包括血管内皮生长因子A、信号转导与转录激活因子3、胱天蛋白酶3、非受体酪氨酸激酶、缺氧诱导因子-1α、表皮生长因子受体等。GO功能富集分析发现生物过程涉及对异物刺激的反应、丝裂原活化蛋白激酶级联的积极调节、跨膜受体蛋白酪氨酸激酶信号通路等方面;细胞组成主要集中在细胞质、细胞核和细胞外泌体等;分子功能主要集中在蛋白质结合、酶活性、脱氧核糖核酸结合等。KEGG通路主要涉及癌症、磷脂酰肌醇-3-激酶/蛋白激酶B、催乳素、缺氧诱导因子-1、乙型肝炎等。结论:猫爪草治疗肝癌具有多成分、多靶点、多途径等特点,为今后的进一步研究提供了理论依据。 |

Objective:To investigate the mechanism of action of Radix Ranunculi Ternati in the treatment of hepatocellular carcinoma (HCC) based on GEO database and network pharmacology.Methods:HERB database,SwissADME,and SwissTargetPrediction were used to obtain the effective constituents and action targets of Radix Ranunculi Ternati.GEO database was used to obtain the differentially expressed genes between the patients with normal liver tissue and the patients with HCC,and GeneCards,DisGeNET,and GEO database were used to obtain the disease targets of HCC and the intersecting genes of the disease and the components.Cytoscape 3.9.0 was used to construct a component-disease network and perform a topological analysis,and David database was used to perform gene ontology (GO) functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis.Results:There were 36 active components of Radix Ranunculi Ternati,with 544 potential targets and 1 950 differentially expressed genes.The core targets included vascular endothelial growth factor A,signal transducer and activator of transcription 3,caspase 3,non-receptor tyrosine kinase,hypoxia-inducible factor-1α,and epidermal growth factor receptor.GO functional enrichment analysis showed that the biological processes involved response to foreign body stimulation,positive regulation of mitogen-activated protein kinase cascades,and the transmembrane receptor protein tyrosine kinase signaling pathway;cellular components mainly included cytoplasm,cell nucleus,and exosome;molecular functions mainly included protein binding,enzyme activity,and deoxyribonucleic acid binding.The KEGG pathways mainly involved cancer,phosphatidylinositol 3-kinase/protein kinase-B,prolactin,hypoxia-inducible factor 1,and hepatitis B.Conclusion:Radix Ranunculi Ternati has the features of multiple components,targets,and pathways in the treatment of HCC,which provides a theoretical basis for subsequent studies. |
