吴茱萸HPLC指纹图谱的建立及质量评价

摘要:
目的:建立吴茱萸高效液相色谱指纹图谱分析方法(HPLC),为其质量评估提供依据。方法:采集3省共22批次吴茱萸药材,通过HPLC法建立指纹图谱,利用《中药色谱指纹图谱相似度评价系统》(2012年版)和IBM SPSS Statistics 26软件进行相似度评价、聚类分析(CA)和主成分分析(PCA),采用1,1-二苯基-2-苦基肼(DPPH)法检测吴茱萸的抗氧化活性。结果:建立了22个批次样品(相似度在0.603~0.992)的HPLC指纹图谱,确定了13个共有峰,并指认出其中6个。以吴茱萸碱为参照峰,计算各共有峰的相对峰面积(RSD为45.29%~112.17%),表明各批次药材中部分共有成分的含量波动较大。相似度结果表明,贵州产地药材相似度更高,其次是湖南,江西产地最低。CA结果显示,22批样品可分为2大类,来源于湖南的3批、江西的6批和贵州的3批聚为第一类,江西剩下的9批和贵州剩下的1批聚为第二类。由PCA结果可知,前3个主成分因子对总方差的累积贡献率达78.164%,可作为吴茱萸整体质量的评价指标。各批次吴茱萸抗氧化活性能力有较大不同,湖南产地批次间自由基清除率差异最小(RSD为9.32%),贵州产地最大(RSD为70.17%),江西居中(RSD为41.94%)。结论:吴茱萸HPLC指纹图谱分析方法简便、快捷,可有效、全面地用于药材的质量评价。贵州产地吴茱萸相似度较高,江西产地质量最佳,湖南产地质量最稳定,因样本量差异,还需积累更多研究数据才能定论。

Abstract:
Objective:To establish the high-performance liquid chromatography (HPLC) fingerprint of Tetradium ruticarpum,and to provide a basis for its quality assessment.Methods:A total of 22 batches of Tetradium ruticarpum were collected from three provinces,and HPLC was performed to establish the fingerprints.The chromatographic fingerprint similarity evaluation system (2012 edition) and IBM SPSS Statistics 26 software were used to perform similarity evaluation,cluster analysis (CA),and principal component analysis (PCA),and the 1,1-diphenyl-2-picrylhydrazyl hydrate method was used to measure the antioxidant activity of Tetradium ruticarpum.Results:The HPLC fingerprints were established for the 22 batches of samples,with a similarity of 0.603~0.992,and a total of 13 common peaks were determined,among which 6 peaks were identified.With evodiamine as the reference peak,the relative peak area was calculated for each common peak,with a relative standard deviation (RSD) of 45.29%~112.17%,suggesting that there were great fluctuations in the content of some common components across different batches of samples.The results of similarity analysis showed the highest degree of similarity in the medicinal material from Guizhou producing areas,followed by that from Hunan producing areas,while the lowest degree of similarity was observed in the medicinal material from Jiangxi producing areas.The results of CA showed that the 22 batches of samples were clustered into 2 categories,with 3 batches from Hunan,6 batches from Jiangxi,and 3 batches from Guizhou as the first category and the remaining 9 batches from Jiangxi and the remaining 1 batch from Guizhou as the second category.The results of PCA showed that the top three principal components had a cumulative variance contribution rate of 78.164% and could be used as assessment indices for the overall quality of Tetradium ruticarpum.There was a difference in the antioxidant activity of Tetradium ruticarpum across different batches;the batches from Hunan producing areas showed the least difference in free radical clearance rate (RSD=9.32%),while those from Guizhou producing areas showed the greatest difference (RSD=70.17%),and those from Jiangxi producing areas showed a moderate difference (RSD=41.94%).Conclusion:The HPLC fingerprint analysis of Tetradium ruticarpum is simple and rapid and can be used for the quality assessment of medicinal material.Tetradium ruticarpum from Guizhou producing areas has a relatively high degree of similarity,that from Jiangxi producing areas has the best quality,and that from Hunan producing areas has the most stable quality.Due to the difference in sample size,more research data are needed to achieve a final conclusion.
