基于网络药理学探讨凉血五花汤加味治疗玫瑰痤疮的作用机制

摘要:
目的:基于网络药理学探讨凉血五花汤加味治疗玫瑰痤疮的作用机制。方法:通过TCMSP获取凉血五花汤加味的有效活性成分及其对应靶点,从GeneCards、DrugBank、OMIM、TTD数据库检索玫瑰痤疮靶点,运用Cytoscape 3.7.2软件构建药物-成分-靶点网络图,借用STRING数据库构建药物和疾病交集靶点蛋白质-蛋白质相互作用(PPI)网络,通过DAVID数据库与微生信在线生物信息学分析可视化云平台对核心靶点基因进行基因本体(GO)功能和京都基因与基因组百科全书(KEGG)通路富集分析。结果:从凉血五花汤加味中筛选出42个活性成分,关键成分包括槲皮素、山柰酚、木犀草素、β-胡萝卜素、青蒿素等,预测靶点222个,药物疾病共同靶点45个,核心靶点为白细胞介素-6(IL-6)、表皮生长因子受体(EGFR)、肿瘤坏死因子(TNF)、半胱氨酸蛋白酶-3(CASP3)、基质金属蛋白酶-9(MMP9)等;GO功能和KEGG通路富集分析筛选出114条信号通路,主要涉及TNF、白细胞介素-17(IL-17)、高级糖基化终末产物-受体(AGE-RAGE)、NOD样受体(NLR)信号通路等。结论:凉血五花汤加味可能基于多成分、多靶点、多通路的特点,相互协同发挥对玫瑰痤疮的治疗作用。

Abstract:
Objective:To investigate the mechanism of action of Jiawei Liangxue Wuhua decoction in the treatment of acne rosacea based on network pharmacology.Methods:TCMSP was used to obtain the active components of Jiawei Liangxue Wuhua decoction and their corresponding targets,and GeneCards,DrugBank,OMIM,and TTD databases were used to obtain the targets of acne rosacea.Cytoscape 3.7.2 was used to construct a drug-component-target network;STRING database was used to construct a protein-protein interaction network for the intersecting targets of the drug and the disease;DAVID database and Weishengxin Online Bioinformatics Visualization Cloud Platform were used to perform gene ontology (GO) functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis.Results:A total of 42 active components were obtained for Jiawei Liangxue Wuhua decoction,with the key components including quercetin,kaempferol,luteolin,β-carotene,and artemisinin,and there were 222 predicted targets and 45 intersecting targets of the drug and the disease,with the key targets including interleukin-6,epidermal growth factor receptor,tumor necrosis factor (TNF),CASP3,and matrix metalloproteinase-9.The GO functional enrichment analysis and the KEGG pathway enrichment analysis obtained 114 signaling pathways,mainly including the TNF,interleukin-17,AGE-RAGE,and NOD-like receptor signaling pathways.Conclusion:Jiawei Liangxue Wuhua decoction exerts a therapeutic effect on acne rosacea through the synergistic effect of multiple components,targets,and pathways.
