目的:基于网络药理学探讨固表防感汤发挥免疫调节的作用机制。方法:在TCMSP平台中检索黄芪、大枣的活性成分及其可能的作用靶点,在OMIM及GeneCards中设置关键词检索免疫调节的关键靶点,将两者检索所得靶点取交集得到固表防感汤的免疫调节靶点,将交集靶点导入STRING数据库中进行蛋白质-蛋白质相互作用网络分析,使用 Metascape数据库对交集靶点进行基因本体(GO)功能、京都基因与基因组百科全书(KEGG)通路富集分析,通过Cytoscape3.9软件分析得到药物成分-疾病靶点-通路网络图。结果:固表防感汤中进行免疫调节的主要关键活性成分为槲皮素、β-胡萝卜素、β-谷甾醇、山柰酚等,其作用于人类V-Rel网状内皮增生病毒癌基因同源物A、肿瘤坏死因子、白细胞介素-6、白细胞介素-10等相关靶点。GO功能富集分析结果显示,固表防感汤在生物过程上通过脂多糖类合成反应、细胞迁移正向调控、造血相关及细胞凋亡等通路来发挥免疫调节作用;分子功能则体现在细胞因子受体结合、激酶结合及活性调节、血红素结合、蛋白域结合等方面。细胞组分主要分布在脂筏、囊泡腔、转录调节复合物、细胞质膜外侧、线粒体外膜等。KEGG通路富集分析结果显示,其通过代谢相关、免疫细胞、细胞因子、肿瘤相关方面的信号通路等多条信号通路发挥免疫调节作用。结论:固表防感汤中多种活性成分通过代谢相关、免疫细胞、细胞因子、肿瘤相关方面的多条信号通路发挥免疫调节作用,为该方在临床上的进一步使用提供了理论基础。 |

Abstract: |
Objective:To investigate the mechanism of action of Gubiao Fanggan decoction in exerting an immunoregulatory effect based on network pharmacology.Methods:TCMSP platform was used to obtain the active components of Astragalus membranaceus and jujube and their possible action targets,and related keywords were used to search for the key targets of immunoregulation in OMIM and GeneCards;intersecting targets were obtained for these two groups of targets to obtain the immunoregulatory targets of Gubiao Fanggan decoction.The intersecting targets were imported into STRING database to perform a protein-protein interaction network analysis,and Metascape database was used to perform gene ontology (GO) functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis of the intersecting genes.Cytoscape 3.9 software was used to construct the drug component-disease target-pathway network.Results:The main key active components of Gubiao Fanggan decoction for immunoregulation included quercetin,beta-carotene,beta-sitosterol,and kaempferol,which acted on the targets such as human V-Rel reticuloendotheliosis viral oncogene homolog A,tumor necrosis factor,interleukin-6,and interleukin-10.GO functional enrichment analysis showed that Gubiao Fanggan decoction played an immunoregulatory role through the pathways such as lipopolysaccharide synthetic reaction,positive regulation of cell migration,hematopoiesis,and cell apoptosis,and its molecular function was reflected in the aspects of cytokine-receptor binding,kinase binding and activity regulation,heme binding,and protein domain binding.Cellular components were mainly distributed in lipid raft,vesicles,transcriptional regulation complex,outer side of plasma membrane,and outer mitochondrial membrane.KEGG pathway enrichment analysis showed that Gubiao Fanggan decoction played an immunoregulatory role through multiple signaling pathways associated with metabolism,immune cells,cytokines,and tumor.Conclusion:The multiple active components in Gubiao Fanggan decoction play an immunoregulatory role through multiple signaling pathways associated with metabolism,immune cells,cytokines,and tumor,which provides a theoretical basis for further application of this prescription in clinical practice. |
