目的:利用网络药理学构建七味白术散药效成分、腹泻、靶点及通路间的网络关系,探讨七味白术散治疗腹泻的作用机制。方法:利用中药系统药理分析平台查询筛选七味白术散的药效成分及对应靶点,从GeneCards数据库获取腹泻相关靶点,使用在线软件Venny 2.1筛选药物与疾病作用的交集靶点;基于David数据库对潜在靶标进行基因本体(GO)功能及京都基因与基因组百科全书(KEGG)通路富集分析。利用STRING在线数据库构建蛋白相互作用网络;用Cytoscape软件绘制活性成分-疾病靶点关系网络。结果:七味白术散的药效成分有125种,以黄酮类成分为主;其治疗腹泻的关键靶点有丝氨酸/苏氨酸蛋白激酶(AKT1)、血管内皮生长因子A(VEGFA)、白细胞介素-6(IL-6)、肿瘤抑制因子p53(TP53)等。GO富集分析后得到与生物过程(BP)相关的有药物响应、基因表达调控、细胞凋亡调控和RNA聚合酶II启动子的转录调节;KEGG通路主要涉及乙型肝炎、结直肠癌、胰腺癌、锥虫病、肿瘤坏死因子(TNF)信号通路、膀胱癌等。结论:七味白术散中的主要成分为黄酮类,通过抑制炎症反应及促进肠黏膜修复等途径起到抗腹泻的作用。 |

Mechanism of action of Qiwei Baizhu powder in treatment of diarrhea:An analysis based on network pharmacology |

Abstract: |
Objective:To investigate the mechanism of action of Qiwei Baizhu powder in the treatment of diarrhea by establishing the effective constituents of Qiwei Baizhu powder and the network relationship between diarrhea,targets,and pathways based on network pharmacology.Methods:Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform was used to screen out the effective constituents of Qiwei Baizhu powder and their corresponding targets,and GeneCards database was used to obtain the targets associated with diarrhea.The online software Venny 2.1 was used to screen out the intersecting targets between the drugs and the disease,and David database was used to perform gene ontology (GO) functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis.STRING online database was used to construct a protein-protein interaction network,and Cytoscape software was used to plot the active component-disease target network.Results:A total of 125 effective constituents were screened out for Qiwei Baizhu powder,most of which were flavonoids,and the key targets for Qiwei Baizhu powder in the treatment of diarrhea included AKT1,vascular endothelial growth factor A,interleukin-6,and TP53.GO enrichment analysis showed that drug response,gene expression and regulation,cell apoptosis regulation,and transcriptional regulation of RNA polymerase II promoter were associated with biological processes,and KEGG pathways mainly involved hepatitis B,colorectal cancer,pancreatic cancer,trypanosomiasis,tumor necrosis factor signaling pathway,and bladder cancer.Conclusion:flavonoids are the main components of Qiwei Baizhu powder,which exerts a therapeutic effect on diarrhea by inhibiting inflammatory response and promoting the repair of intestinal mucosa. |
