天降血栓通丸对ApoE-/-小鼠动脉粥样硬化的干预作用及其机制研究

摘要:
目的:探讨天降血栓通丸对Apo-/-小鼠动脉粥样硬化的干预作用及其机制。方法:将30只动脉粥样硬化建模成功的ApoE-/-小鼠随机分为模型组、阿托伐他汀组和天降血栓通丸组,每组各10只,分别给予0.9%氯化钠注射液、阿托伐他汀、天降血栓通丸药液灌胃8周。10只C57BL/6小鼠予普通饲料喂养,作为对照组,灌胃0.9%氯化钠注射液。干预结束后,检测血清总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、单核细胞趋化因子-1(MCP-1)、细胞间黏附分子-1(ICAM-1)、血管细胞黏附分子-1(VCAM-1)水平,检测主动脉组织c-Jun氨基末端活化蛋白激酶(JNK)、丝裂原活化蛋白激酶(p38MAPK)mRNA的表达。结果:与模型组比较,阿托伐他汀组和天降血栓通丸组血清TG、TC、LDL-C、MCP-1、ICAM-1、VCAM-1水平及主动脉组织JNK mRNA、p38MAPK mRNA表达均降低,差异均有统计学意义(P<0.05);与对照组比较,模型组血清TG、TC、LDL-C、MCP-1、ICAM-1、VCAM-1水平及主动脉组织JNK mRNA、p38MAPK mRNA表达水平均升高,差异均有统计学意义(P<0.05)。结论:天降血栓通丸具有防治动脉粥样硬化的作用,其机制可能与抑制JNK/p38 MAPK信号,下调MCP-1、ICAM-1、VCAM-1表达,减轻炎症反应有关。

Abstract:
Objective:To investigate the interventional effect of Tianjiang Xueshuantong pills on atherosclerosis in ApoE-/- mice and its mechanism.Methods:A total of 30 ApoE-/- mice with successful atherosclerosis modeling model,which were randomly divided into model group,atorvastatin group,and Tianjiang Xueshuantong pill group,with 10 mice in each group.The above three groups were given 0.9% sodium chloride injection,atorvastatin solution,and Tianjiang Xueshuantong pill solution,respectively,by gavage for 8 weeks.A total of 10 C57BL/6 mice fed with normal diet were selected as control group and were given 0.9% sodium chloride injection by gavage.After intervention,the serum levels of total cholesterol (TC),triglyceride (TG),low-density lipoprotein cholesterol (LDL-C),high-density lipoprotein cholesterol,monocyte chemoattractant protein-1(MCP-1),intercellular adhesion molecule-1 (ICAM-1),and vascular cell adhesion molecule-1 (VCAM-1) were measured,as well as the mRNA expression of c-Jun N-terminal kinase (JNK) and p38 mitogen-activated protein kinase (p38 MAPK) in aortic tissue.Results:Compared with the model group,the atorvastatin group and the Tianjiang Xueshuantong pill group had significant reductions in the serum levels of TG,TC,LDL-C,MCP-1,ICAM-1,and VCAM-1 and the mRNA expression of JNK and p38 MAPK in aortic tissue (P<0.05).Compared with the control group,the model group had significant increases in the serum levels of TG,TC,LDL-C,MCP-1,ICAM-1,and VCAM-1 and the mRNA expression of JNK and p38 MAPK in aortic tissue (P<0.05).Conclusion:Tianjiang Xueshuantong pills have a certain effect in the prevention and treatment of atherosclerosis,possibly by inhibiting the JNK/p38 MAPK signal,downregulating the expression of MCP-1,ICAM-1,and VCAM-1,and alleviating inflammatory response.
