基于网络药理学探讨“黄芪-桂枝”治疗慢性心力衰竭的作用机制

摘要:
目的:运用网络药理学技术探讨“黄芪-桂枝”治疗慢性心力衰竭(CHF)的作用机制。方法:通过中药系统药理学数据库与分析平台(TCMSP)筛选出“黄芪-桂枝”中潜在活性化合物及其潜在作用靶点,采用Gene Cards、OMIM数据库获得相关疾病靶点,通过String数据库构建关键靶点蛋白相互作用网络,使用Metascape在线工具对共同靶点基因进行GO功能和KEGG通路富集分析。并将相关数据导入Cytoscape 3.6.1软件进行可视化分析,构建“活性成分-CHF-靶点”网络模型和“靶点-GO-KEGG”网络模型。结果:筛选出27个主要活性成分,共涉及207个中药有效成分和175个中药-疾病关键靶点。“黄芪-桂枝”主要通过调控丝氨酸/苏氨酸蛋白激酶1(AKT1)、丝裂原活化蛋白激酶-1(MAPK1)、白细胞介素-6(IL-6)、转录因子Ap-1(JUN)、肿瘤坏死因子(TNF)等蛋白,以及参与癌症途径、乙型肝炎信号通路、磷脂酰肌醇3-激酶(PI3K)-蛋白激酶B(Akt)信号通路、癌症蛋白多糖通路、人类T淋巴细胞瘤病毒Ⅰ型(HTLV-I)感染通路等发挥治疗CHF的作用。结论:本研究揭示了“黄芪-桂枝”组分中的有效化合物以多靶点、多通路、多途径治疗CHF的作用机制,为临床研究提供了新思路。

Abstract:
Objective:To investigate the mechanism of action of Astragalus membranaceus-Ramulus Cinnamomi in the treatment of chronic heart failure (CHF) based on network pharmacology.Methods:The Traditional Chinese Medicine Systems Pharmacology platform was used to screen out the potential active components of Astragalus membranaceus-Ramulus Cinnamomi and their potential action targets;the Gene Cards and OMIM databases were used to obtain the disease targets;the String database was used to construct a protein-protein interaction network of key targets;the Metascape online tool was used to perform GO enrichment analysis and KEGG pathway enrichment analysis.Related data were imported into Cytoscape 3.6.1 for visual analysis,and an “active component-CHF-target” network model and a “target-GO-KEGG” network model were established.Results:A total of 27 main active components were screened out,which involved 207 effective constituents and 175 key traditional Chinese medicine-disease targets.Astragalus membranaceus-Ramulus Cinnamomi exerted a therapeutic effect on CHF mainly by regulating the proteins including RAC-alpha serine/threonine-protein kinase,mitogen-activated protein kinase 1,interleukin-6,transcription factor Ap-1,and tumor necrosis factor and participating in the cancer pathway,the hepatitis B signaling pathway,the phosphoinositide 3-kinase-protein kinase B signaling pathway,the cancer proteoglycan pathway,and the HTLV-I infection pathway.Conclusion:This study revealed the mechanism of action of the effective compounds in the "Astragalus-Guizhi" component in the treatment of CHF with multiple targets, multiple pathways, and multiple ways, providing a new idea for clinical research.
