| 摘要: |
| 目的：研究咳喘宁对哮喘豚鼠白细胞介素（IL-6）和肿瘤坏死因子（TNF-α）的调节作用。方法：将60只豚鼠随机分为空白组10只、造模组50只。利用呼吸道合胞病毒（RSV）感染豚鼠建立动物模型,加鸡卵白蛋白（OVA）诱导建立哮喘模型。造模组成功后再将其随机分为模型组,咳喘宁高、中、低剂量组,西药组,每组各10只。咳喘宁高、中、低剂量组分别给予不同剂量的咳喘宁口服液,西药组给予地塞米松+利巴韦林,空白组、模型组给予0.9%氯化钠注射液。经治疗7 d后,应用HE染色法检测咳喘宁对RSV哮喘豚鼠模型的治疗效果;收集支气管肺泡灌洗液（BALF）并采集心脏血,用酶联免疫吸附（ELISA）法分析血清、BALF中IL-6和TNF-α的含量。结果：血清、BALF中IL-6、TNF-α含量,模型组与空白组比较,差异有统计学意义（P＜0.05）,说明造模成功。咳喘宁高、中、低剂量组及西药组与模型组比较,差异均有统计学意义（P＜0.05）。咳喘宁高剂量组与西药组比较,差异亦有统计学意义（P<0.05）。结论：咳喘宁可以降低炎性因子水平,控制气道炎症,调节IL-6和TNF-α含量,改善哮喘模型豚鼠的肺组织病理变化。 |

| Regulatory effect of Kechuanning on interleukin-6 and tumor necrosis factor-α in guinea pigs with asthma |

| Objective：To investigate the regulatory effect of Kechuanning on interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) in guinea pigs with asthma.Methods：A total of 60 guinea pigs were randomly divided into blank group with 10 guinea pigs and modeling group with 50 guinea pigs.The guinea pigs were infected with respiratory syncytial virus (RSV) to establish an animal model,and ovalbumin induction was performed to establish a model of asthma.After modeling,the guinea pigs in the modeling group were further divided into model group,high-,middle-,and low-dose Kechuanning groups,and Western medicine group,with 10 guinea pigs in each group.The guinea pigs in the high-,middle-,and low-dose Kechuanning groups were given different doses of Kechuanning oral liquid,those in the Western medicine group were given dexamethasone+ribavirin,and those in the blank group and the model group were given 0.9% sodium chloride injection.After 7 days of treatment,HE staining was used to investigate the therapeutic effect of Kechuanning on guinea pigs with RSV-induced asthma;bronchoalveolar lavage fluid (BALF) and heart blood were collected,and ELISA was used to measure the content of IL-6 and TNF-α in serum and BALF.Results：There were significant differences in the content of IL-6 and TNF-α in serum and BALF between the model group and the blank group (P＜0.05),suggesting that the model was successfully established.There were significant differences between the high-,middle-,and low-dose Kechuanning groups and the model group and between the Western medicine group and the model group (P＜0.05),and there were also significant differences between the high-dose Kechuanning group and the Western medicine group (P＜0.05).Conclusion：In guinea pigs with asthma,Kechuanning can reduce the levels of inflammatory factors,control airway inflammation,regulate the content of IL-6 and TNF-α,and improve pathological changes in lungs. |
