加味丹参饮通过PTEN/PI3K/Akt信号通路抑制IRI作用的研究

摘要:
目的:探讨加味丹参饮预处理通过抑瘤基因/磷脂酰肌醇-3激酶/蛋白激酶B(PTEN/PI3K/Akt)信号通路对大鼠心肌缺血再灌注损伤(IRI)的保护作用及机制。方法:将75只雄性远交群(SD)大鼠随机分为5组:假手术组、模型组、加味丹参饮组、加味丹参饮+PI3K/Akt通路抑制剂(LY294002)组、LY294002组,采用结扎大鼠冠状动脉左前降支30min后再灌注60min的方法制备IRI模型。采用酶联免疫吸附测定(ELISA)法检测各组大鼠心肌肌钙蛋白I(cTnI)表达;取心肌梗死边缘区心肌组织,采用脱氧核糖核苷酸末端转移酶介导的缺口末端标记(TUNEL)法检测心肌凋亡指数,采用免疫组化法检测各组大鼠心肌组织Akt、PTEN的表达。结果:与假手术组比较,模型组cTnI水平显著升高,大鼠心肌凋亡指数增加,PTEN、Akt表达均显著升高(P<0.05);与模型组比较,加味丹参饮组cTnI水平均显著降低,Akt表达升高,心肌凋亡指数和PETN表达降低(P<0.05);与加味丹参饮组比较,加入抑制剂后,cTnI水平均明显降低,Akt表达降低,PTEN表达升高(P<0.05)。结论:大鼠心肌缺血再灌注时,加味丹参饮可以通过降低PTEN表达,激活PI3K/Akt信号通路,抑制大鼠心肌细胞的凋亡,从而减轻心肌损伤,保护心肌。

Abstract:
Objective:To investigate the protective effect of Jiawei Danshen decoction pretreatment against myocardial ischemia-reperfusion injury (IRI) in rats via the PTEN/PI3K/Akt signaling pathway and related mechanisms.Methods:A total of 75 male outbred Sprague-Dawley rats were randomly divided into sham-operation group,model group,Jiawei Danshen decoction group,Jiawei Danshen decoction+PI3K/Akt pathway inhibitor (LY294002) group,and LY294002 group.A rat model of IRI was established by ligation of the left anterior descending coronary artery,followed by 60-minute reperfusion after 30 minutes.ELISA was used to measure the expression of cardiac troponin I (cTnI).Myocardial tissue was collected from the infarct border zone;terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling was used to measure myocardial apoptotic index and immunohistochemistry was used to measure the expression of Akt and PTEN in myocardial tissue.Results:Compared with the sham-operation group,the model group had significant increases in cTnI,myocardial apoptotic index,and expression of PTEN and Akt (P<0.05).Compared with the model group,the Jiawei Danshen decoction group had a significant reduction in cTnI,a significant increase in the expression of Akt,and significant reductions in myocardial apoptotic index and PTEN (P<0.05).Compared with the Jiawei Danshen decoction,the Jiawei Danshen decoction+LY294002 group had significant reductions in cTnI and Akt and a significant increase in PTEN (P<0.05).Conclusion:In a rat model of myocardial IRI,Jiawei Danshen decoction can alleviate myocardial injury and protect the myocardium by reducing the expression of PTEN,activating the PI3K/Akt signaling pathway,and inhibiting the apoptosis of cardiomyocytes.
