目的:研究苦参汤对皮肤溃疡大鼠伤口愈合的影响。方法:通过皮下注射75%冰醋酸诱导大鼠皮肤溃疡模型,将模型大鼠分为5组,G1采用蒸馏水干预,G2~5分别采用苦参汤低、中、高剂量及0.02%高锰酸钾溶液(PP)进行局部治疗,连续7d。治疗后对各组溃疡面积、溃疡评分和组织学评估,通过免疫组织化学分析检测前炎性细胞因子前列腺素E2(PGE-2)和白细胞介素-8(IL-8)的表达。结果:苦参汤对皮肤溃疡大鼠创口愈合有促进作用,中剂量和高剂量苦参汤治疗的大鼠溃疡愈合更佳(P<0.05),低剂量苦参汤治疗的肉眼可见溃疡评分显著降低(P<0.01),PP组在降低溃疡面积和肉眼可见的溃疡评分方面,差异无统计学意义(P>0.05);所有剂量的苦参汤和PP均改善了大鼠诱导的皮肤溃疡的组织学损伤;高剂量苦参汤和PP使大鼠皮肤溃疡组织中的PGE-2和IL-8表达显著降低(P<0.01);各组大鼠均未发现与治疗有关的毒性。结论:苦参汤可促进皮肤溃疡大鼠的伤口愈合,其对PGE-2和IL-8的抑制是增强伤口愈合的可能作用机制。 |

Effect of Kushen decoction on wound healing in rats with skin ulcer |

Abstract: |
Objective:To investigate the effect of Kushen decoction on wound healing in rats with skin ulcer.Methods:Subcutaneous injection of 75% glacial acetic acid was given to establish a rat model of skin ulcer,and then the model rats were divided into groups 1-5.The rats in group 1 were given distilled water,and those in groups 2~5 were given local treatment with low-,middle-,and high-dose Kushen decoction and 0.02% potassium permanganate (PP) solution,respectively.The course of treatment was 7 consecutive days.Ulcer area,ulcer score,and histology were evaluated after treatment,and immunohistochemistry was used to measure the expression of the proinflammatory cytokines prostaglandin E2(PGE2) and interleukin-8 (IL-8).Results:Kushen decoction promoted wound healing in rats with skin ulcer,and middle- and high-dose Kushen decoction had a better effect (P<0.05),while low-dose Kushen decoction significantly reduced macroscopic ulcer score (P<0.01).The group treated with PP had reductions in ulcer area and macroscopic ulcer score (P>0.05).Low-,middle-,and high-dose Kushen decoction and PP improved the histological injury of skin ulcer.High-dose Kushen decoction and PP significantly reduced the expression of PGE-2 and IL-8 in skin ulcer tissue (P<0.01).No treatment-related toxicity was observed.Conclusion:Kushen decoction can promote wound healing in rats with skin ulcer,possibly by inhibiting PGE-2 and IL-8. |
