目的:探讨强骨颗粒对骨质疏松大鼠胫骨组织中骨保护素(OPG)、核因子κB受体活化因子配基(RANKL)表达的影响,并分析其改善骨质疏松的作用机制。方法:建立骨质疏松大鼠模型,随机分为对照组、假手术组、模型组、强骨颗粒组、骨疏康颗粒组和尼尔雌醇片组,每组各6只,予相应药物灌胃,对照组、假手术组、模型组予以等剂量0.9%氯化钠注射液灌胃。分别于给药后第3、6周每组随机抽取2只处死,取患处组织标本,采用染色后视野下阳性表达部位的累积光密度和视野下样品面积的比值法测定大鼠骨组织中OPG、RANKL的含量,对所得数据进行统计学分析。结果:强骨颗粒组大鼠在第3、6周末患处骨组织标本中OPG、RANKL表达均高于其他组(P<0.05)。结论:强骨颗粒可促进骨质疏松骨组织中OPG、RANKL的表达,改善骨质疏松。 |

Effect of Qianggu granules on the expression of osteoprotegerin and receptor activator of nuclear factor-kappa B ligand in tibial tissue in rats with osteoporosis |

Objective:To investigate the effect of Qianggu granules on the expression of osteoprotegerin (OPG) and receptor activator of nuclear factor-kappa B ligand (RANKL) in tibial tissue in rats with osteoporosis,as well as the mechanism of action of Qianggu granules in improving osteoporosis.Methods:A rat model of osteoporosis was established.The rats were randomly divided into control group,sham-operation group,model group,Qianggu granules group,Gushukang granules group,and nilestriol tablets group,with 6 rats in each group.The corresponding drug was given by gavage,and the rats in the control group,the sham-operation group,and the model group were given an equal dose of 0.9% sodium chloride injection by gavage.Two rats were randomly selected from each group and sacrificed at weeks 3 and 6 of administration,and tissue specimens of the lesion were collected.The content of OPG and RANKL in bone tissue was measured by integrated optical density of the positive expression site and area ratio in the visual field after staining,and a statistical analysis was performed on the obtained data.Results:At the end of weeks 3 and 6,the Qianggu granules group had significantly higher expression of OPG and RANKL in bone tissue of the lesion than the other groups (P<0.05).Conclusion:Qianggu granules can promote the expression of OPG and RANKL in bone tissue and thus improve osteoporosis. |
