石柑子不同提取部位对RAW264.7细胞炎症的影响

摘要:
目的:观察石柑子不同提取部位对脂多糖(LPS)诱导的小鼠单核巨噬细胞RAW264.7细胞炎症的影响。方法:用不同质量浓度的石柑子石油醚振摇提取部分、三氯甲烷振摇提取部分、乙酸乙酯振摇提取部分、正丁醇振摇提取部分以及水提取部分和阳性药地塞米松处理LPS刺激的RAW264.7细胞。分别运用噻唑蓝(MTT)法检测细胞活性,RT-qPCR法检测iNOS细胞和COX-2细胞的mRNA表达,Western blot法检测iNOS细胞和COX-2细胞的蛋白表达。结果:石柑子石油醚提取部位、三氯甲烷提取部位、乙酸乙酯提取部位、正丁醇提取部位和水提取部位作用RAW264.7细胞的无毒剂量分别为25、5、10、50、25μg/ml;LPS的用药质量浓度为0.5μg/ml;地塞米松的用药质量浓度为0.4μg/ml;石油醚提取部位25μg/ml和地塞米松能下调iNOS mRNA表达且抑制iNOS的蛋白表达;三氯甲烷提取部位5μg/ml和地塞米松能抑制COX-2的蛋白表达,并且下调COX-2 mRNA的表达。结论:石油醚振摇提取部分和三氯甲烷振摇提取部分对通过LPS诱导的RAW264.7细胞炎症具有抑制作用,其作用机制可能与抑制iNOS和COX-2细胞基因核蛋白的表达有关。

Abstract:
Objective:To observe the effects of Pothos chinensis extracts in different solvents on lipopolysaccharides(LPS)-induced cellular inflammation in mouse RAW264.7 macrophages.Methods:The LPS-treated RAW264.7 cells were treated with the positive control dexamethasone and different mass concentrations of Pothos chinensis extracts in petroleum ether,chloroform,ethyl acetate,n-butanol,and water.Cell viability was evaluated by MTT assay.RT-qPCR and Western blot were used to determine the mRNA and protein expression of iNOS and COX-2 in cells,respectively.Results:the non-toxic doses of Pothos chinensis extracts in petroleum ether,chloroform,ethyl acetate,n-butanol,and water for RAW264.7 cells were 25,5,10,50,and 25μg/ml,respectively.The concentrations of LPS and dexamethasone used to treat cells were 0.5 and 0.4μg/ml.The Pothos chinensis extract in petroleum ether at a concentration of 25μg/ml and dexamethasone reduced the mRNA and protein expression of iNOS in cells.The Pothos chinensis extract in chloroform at a concentration of 5μg/ml and dexamethasone reduced the mRNA and protein expression of COX-2 in cells.Conclusion:The Pothos chinensis extracts in petroleum ether and chloroform can inhibit the LPS-induced cellular inflammation in RAW264.7 cells,probably by reducing the expression of iNOS and COX-2.
