目的:探讨芒果苷对高脂肪/高果糖饮食诱导大鼠代谢综合征(MS)的影响。方法:将60只大鼠随机分为正常组,模型组,阳性对照组,芒果苷高、中、低剂量组,每组各10只。每天饲喂17%脂肪/17%果糖饲料+20%果糖水,构建MS模型,造模8周。造模成功后阳性对照组灌胃二甲双胍50mg/kg,芒果苷高、中、低剂量组分别灌胃40、20、10mg/kg,正常组和模型组灌胃等体积0.5%羧甲基纤维素钠,连续给药8周。观察各组的体质量、内脏脂肪质量、脂体比及血清总胆固醇、三酰甘油、空腹血糖、胰岛素水平、胰岛素抵抗指数(IRI)、脂肪细胞因子(脂联素、抵抗素)水平。结果:与模型组比较,芒果苷高、中、低剂量组的体质量、内脏脂肪质量、脂体比、血清总胆固醇、三酰甘油、空腹血糖、胰岛素及抵抗素水平下降(P<0.01),芒果苷高、中剂量组IRI降低(P<0.05)。结论:芒果苷通过调控糖脂代谢、脂肪细胞因子、脂肪堆积、胰岛素抵抗等途径干预大鼠MS的病理进程。 |

Effect of mangiferin on rats with metabolic syndrome induced by high-fat/high-fructose diet |

Objective:To investigate the effect of mangiferin on rats with metabolic syndrome (MS) induced by high-fat/high-fructose diet.Methods:A total of 60 rats were randomly divided into normal group,model group,positive control group,and high-,middle-,and low-dose mangiferin groups,with 10 rats in each group.A model of MS was established by feeding 17% fat/17% fructose diet and 20% fructose solution for 8 weeks of modeling.After successful modeling,the rats in the positive control group were given metformin 50 mg/kg by gavage,those in the high-,middle-,and low-dose mangiferin groups were given mangiferin at a dose of 40,20,and 10 mg/kg,respectively,by gavage,and those in the normal group and the model group were given an equal volume of 0.5% sodium carboxymethyl cellulose by gavage,for 8 consecutive weeks.Each group was observed in terms of body weight,visceral fat mass,fat/body weight ratio,the serum levels of total cholesterol/triglyceride/fasting blood glucose/insulin,insulin resistance index (IRI),and the levels of adipokines (adiponectin and resistin).Results:Compared with the model group,the high-,middle-,and low-dose mangiferin groups had significant reductions in body weight,visceral fat mass,fat/body weight ratio,and the serum levels of total cholesterol,triglyceride,fasting blood glucose,insulin,and resistin (P<0.01),and the high- and middle-dose mangiferin groups had a significant reduction in IRI (P<0.05).Conclusion:Mangiferin intervenes against the pathological process of MS in rats by regulating glucose and lipid metabolism,adipokines,fat accumulation,and insulin resistance. |
