肠道湿热型泄泻模型小鼠肠道黏膜微生物活度的动态变化研究

摘要:
目的:观察肠道湿热型泄泻模型小鼠肠道黏膜微生物活度的动态变化,为肠道湿热型泄泻的发生及预防提供实验依据。方法:以“高糖高脂+高温高湿+白酒+冰水”的方法制备肠道湿热型泄泻小鼠模型,分别在造模第0、4、8、11、15 天无菌采集小鼠小肠前、中、后段黏膜,采用荧光素二乙酸酯法测定样品中微生物活度。结果:与正常组相比,肠道湿热型泄泻造模过程中,高糖高脂喂养小鼠肠道前、中、后段黏膜微生物活度显著降低(P<0.01),但在高温高湿造模阶段后期又有不同程度的回升,且在造模第11、15天模型组小鼠肠道后段黏膜微生物活度高于正常组,组间比较,差异有统计学意义(P<0.01)。结论:肠道湿热型泄泻的发生可能与肠道黏膜生物总量的改变有关,合理的饮食结构及适宜的生活环境对疾病的预防具有重要意义。

Abstract:
Objective:To observe the dynamic changes of intestinal mucosal microbial activity in a mouse model of diarrhea with intestinal dampness-heat syndrome,and to provide an experimental basis for preventing diarrhea with intestinal dampness-heat syndrome.Methods:A mouse model of diarrhea with intestinal dampness-heat syndrome was established by high-glucose high-fat diet,high temperature and humidity,liquor,and ice water.Sterile techniques were used to collect the mucosa of the anterior,middle,and posterior segments of the small intestine on days 0,4,8,11 and 15 respectively,and the fluorescein diacetate method were used to measure microbial activity.Results:Compared with the normal group,there was a significant reduction in microbial activity in the mucosa of the anterior,middle,and posterior segments of the small intestine in the mice fed with a high-glucose high-fat diet during the establishment of the model of diarrhea with intestinal dampness-heat syndrome (P<0.01),while varying degrees of increase were observed in the later stage of modeling with high temperature and humidity,and compared with the normal group on days 11 and 15 of modeling,the model group had a significantly higher mucosal microbial activity of the posterior segment of the intestinal tract (P<0.01).Conclusion:The onset of diarrhea with intestinal dampness-heat syndrome might be associated with the change in the total number of intestinal mucosal microbes,and a proper dietary structure and an appropriate living environment are of great significance to the prevention of the disease.
