基于网络药理学研究香附治疗女性青春期后痤疮的作用机制

摘要:
目的:基于网络药理学研究香附治疗女性青春期后痤疮的作用机制。方法:通过TCMSP数据库获取香附的活性成分与相应靶点,采用GeneCard数据库获得与痤疮相关的疾病靶点,使用STRING数据库和Cytoscape 3.7.1软件构建蛋白质-蛋白质相互作用(PPI)网络,应用Bioconductor数据库进行基因本体(GO)功能、京都基因与基因组百科全书(KEGG)通路富集分析。结果:香附的有效活性成分共18种,痤疮相关靶点570个,潜在靶点21个,药物-疾病靶点21个。PPI 网络显示,白细胞介素-6、血管内皮生长因子A、白三醇过氧化物酶体增殖物激活受体γ、表皮生长因子受体、白三烯醇雌激素受体的连接程度最高。GO功能富集分析结果显示,交集基因主要参与了整合素结合、核受体活性、配体激活转录因子活性、ATP酶结合等。KEGG通路主要涉及脂质与动脉粥样硬化、流体剪切应力与动脉粥样硬化、肿瘤坏死因子信号通路、磷脂酰肌醇-3激酶-蛋白激酶B(PI3K-Akt)信号通路等。结论:香附可通过调节多种生物学功能和各项通路治疗女性青春期后痤疮,为临床运用香附治疗女性青春期后痤疮提供了理论支持。

Abstract:
Objective:To investigate the mechanism of action of Rhizoma Cyperi in the treatment of women with post-adolescent acne based on network pharmacology.Methods:TCMSP database was used to obtain the active components of Rhizoma Cyperi and corresponding targets,and GeneCard database was used to obtain the disease targets associated with acne;STRING database and Cytoscape 3.7.1 software were used to construct a protein-protein interaction (PPI) network,and Bioconductor database was used to perform gene ontology (GO) functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis.Results:There were 18 active components of Rhizoma Cyperi,570 targets associated with acne,21 potential targets,and 21 drug-disease targets.The PPI network showed that interleukin-6,vascular endothelial growth factor A,Baisanchun peroxisome proliferator-activated receptor γ,epidermal growth factor receptor,and Baisanxichun estrogen receptor showed the highest degree of connection.GO functional enrichment analysis showed that the intersecting genes were mainly involved in integrin binding,nuclear receptor activity,ligand-activated transcription factor activity,and ATPase binding.KEGG pathways were mainly involved in lipids and atherosclerosis,fluid shear stress and atherosclerosis,the tumor necrosis factor signaling pathway,and the PI3K-Akt signaling pathway.Conclusion:Rhizoma Cyperi exerts a therapeutic effect on post-adolescent acne in women by regulating various biological functions and pathways,which provides a theoretical basis for the clinical application of Rhizoma Cyperi in the treatment of post-adolescent acne in women.
