基于Box-Behnken响应面法的半夏多糖双水相提取工艺优化研究

摘要:
目的:采用Box-Behnken响应面法优化半夏多糖双水相提取工艺。方法:基于单因素试验,选取超声时间、聚乙二醇(PEG)6000用量和 (NH4)22SO4用量为考察指标,将分配系数、相比和提取率的总评归一值作为响应值,以Box-Behnken响应面法优化提取工艺。结果:最佳的半夏多糖双水相提取参数为超声时间44 min,PEG 6000用量1.3 g, (NH4)22SO4用量2.3 g;基于该提取条件测得多糖平均提取率为44.76%,OD值平均值为0.8761,偏差为1.69%。结论:采用PEG 6000- (NH4)22SO4双水相体系分离半夏多糖,其含量高于传统醇沉法,并具有操作简便、成本低、有机溶剂使用少等优点,具有良好的应用前景。

Abstract:
Objective:To optimize the aqueous two-phase extraction process of Pinellia ternate polysaccharides based on the Box-Behnken response surface method.Methods:Based on the single factor experiment and with ultrasound time and the amount of polyethylene glycol (PEG) 6000 and (NH4)22SO4 as evaluation indices,the overall desirability (OD) of distribution coefficient,phase ratio,and extraction rate was used as response,and the Box-Behnken response surface method was used to optimize extraction process.Results:The optimal conditions of aqueous two-phase extraction process for polysaccharides from Pinellia ternate were as follows:Ultrasound time of 44 minutes,1.3 g of PEG 6000,and 2.3 g of (NH4)22SO4.Under such conditions,the mean extraction rate of polysaccharides was 44.76% and the mean OD was 0.8761,with a deviation of 1.69%.Conclusion:Compared with the conventional alcohol deposition method,the PEG 6000- NH4)22SO4 aqueous two-phase extraction process yields a higher content of polysaccharides from Pinellia ternate and has the advantages of simple and convenient operation,low cost,and a small amount of organic solvent used,and therefore,it holds promise for future application.
