益肾健骨颗粒对绝经后骨质疏松大鼠骨组织Wnt/β-catenin通路的影响

摘要:
目的:探讨益肾健骨颗粒对绝经后骨质疏松(PMO)大鼠骨组织Wnt通路LRP5、Wnt3a、β-catenin蛋白表达的影响。方法:将40只大鼠随机分为假手术组、模型组、实验组、阳性对照组,每组各10只。假手术组摘除卵巢周围少量脂肪组织,其余组均切除双侧卵巢以建立PMO模型。12周后测定大鼠双侧股骨骨密度以及骨质疏松骨组织LRP5、Wnt3a、β-catenin蛋白表达水平。结果:与假手术组比较,其余3组骨密度值均下降(P<0.05),说明造模成功;与模型组比较,实验组、阳性对照组骨密度值和LRP5、Wnt3a、β-catenin的蛋白表达水平均升高(P<0.05)。结论:益肾健骨颗粒通过Wnt信号通路促进去卵巢大鼠成骨细胞增殖而改善PMO,效果明显。

Abstract:
Objective:To investigate the effect of Yishen Jiangu granules on the protein expression of LRP5,Wnt3a,and β-catenin of the Wnt/β-catenin pathway in bone tissue of rats with osteoporosis.Methods:A total of 40 rats were randomly divided into sham-operation group,model group,experimental group,and positive control group,with 10 rats in each group.For the rats in the sham-operation group,a small amount of adipose tissue was removed around the ovary,and for those in the other three groups,both ovaries were resected to establish a model of osteoporosis.Bone mineral density of bilateral femurs was measured after 12 weeks,as well as the protein expression levels of LRP5,Wnt3a,and β-catenin in osteoporotic bone tissue.Results:Compared with the sham-operation group,the other three groups had a significant reduction in bone mineral density and the protein expression levels of LRP5,Wnt3a,β-catemin (P<0.05),suggesting that the model was successfully established.Compared with the model group,the experimental group and the positive control group had significant increases in bone mineral density and the protein expression levels of LRP5,Wnt3a,and β-catenin (P<0.05).Conclusion:Yishen Jiangu granules improve osteoporosis by promoting osteoblast proliferation in ovariectomized rats via the Wnt signaling pathway,with good efficacy.
