基于网络药理学探讨黄芪对肾小球肾炎的作用机制

摘要:
目的:运用网络药理学的方法探讨黄芪治疗肾小球肾炎的作用机制。方法:利用TCMSP平台获取黄芪的活性成分及相关的潜在靶点,通过Gene Cards数据库检索有关肾小球肾炎的相关疾病靶点,对中药-疾病靶点取交集,得到黄芪-肾小球肾炎的药效靶点,并采用Cytoscape3.7.2软件构建黄芪-肾小球肾炎靶点网络;使用String数据库进行蛋白相互作用网络(PPI)的绘制;借助R语言以及Bioconductor数据库对黄芪治疗肾小球肾炎潜在作用靶点进行GO分子功能分析和KEGG通路分析。结果:筛选得到黄芪的20个有效成分及98个中药-疾病共同靶标,GO分子功能显示黄芪治疗肾小球肾炎主要涉及细胞因子受体结合、核激素受体结合、细胞因子活性、受体配体活性、转录因子活性等方面。KEGG通路分析结果显示,黄芪治疗肾小球肾炎核心靶点主要涉及晚期糖基化终末产物-糖基化终末产物受体(AGE-RAGE)信号通路、肿瘤坏死因子(TNF)信号通路、白细胞介素-17(IL-17)信号通路、Toll样受体信号通路、缺氧诱导因子-1(HIF-1)信号通路等。结论:黄芪对于肾小球肾炎的治疗作用是多成分-多靶点-多途径的过程,基于网络药理学对此的研究,为临床应用黄芪治疗肾小球肾炎提供了参考价值。

Abstract:
Objective:To investigate the mechanism of action of Astragalus membranaceus on glomerulonephritis based on network pharmacology. Methods:The TCMSP database was used to obtain the active constituents and potential targets of Astragalus membranaceus,and the Gene Cards database was used to search for the targets of glomerulonephritis. The intersection of traditional Chinese medicine (TCM) targets and disease targets was determined to obtain the drug targets of Astragalus membranaceus-glomerulonephritis,and Cytoscape 3.7.2 was used to construct a network of the Astragalus membranaceus-glomerulonephritis targets. String database was used to establish the protein-protein interaction network,and R Language and Bioconductor database were used to perform GO molecular function analysis and KEGG pathway enrichment analysis of the potential targets of Astragalus membranaceus in the treatment of glomerulonephritis. Results:A total of 20 effective constituents of Astragalus membranaceus and 98 TCM-disease targets were screened out,and the GO molecular function analysis showed that cytokine receptor binding,nuclear hormone receptor binding,cytokine activity,receptor-ligand activity,and transcription factor activity were mainly involved in the treatment of glomerulonephritis with Astragalus membranaceus. The KEGG pathway enrichment analysis showed that the core targets of Astragalus membranaceus in the treatment of glomerulonephritis mainly involved the AGE-RAGE signaling pathway,the tumor necrosis factor signaling pathway,the interleukin-17 signaling pathway,the Toll-like receptor signaling pathway,and the hypoxia-inducible factor-1 signaling pathway. Conclusion:The treatment of glomerulonephritis with Astragalus membranaceus is a process involving multiple constituents,targets,and pathways,and such analysis based on network pharmacology provides a reference for the clinical application of Astragalus membranaceus in the treatment of glomerulonephritis.
