基于网络药理学探讨补阳还五汤抗动脉粥样硬化的作用机制

摘要:
目的:利用网络药理学探讨补阳还五汤抗动脉粥样硬化的作用机制。方法:通过TCMSP数据库检索中药成分、靶点,利用SwissTargetPrediction、PubChem 2个在线网站预测中药成分靶点;通过OMIM、DisGeNE、TTD、DrugBank 4个数据库检索动脉粥样硬化靶点;以VENN图展现补阳还五汤靶点与动脉粥样硬化靶点的交集;利用STRING网站构建PPI网络,筛选出关键靶点;通过Cytoscape3.6.0软件筛选出补阳还五汤主要成分;利用DAVID6.8网站对交集靶点精细GO功能分析和KEGG通路富集分析。结果:补阳还五汤靶点与动脉粥样硬化靶点的交集靶点有94个,补阳还五汤活性成分62个,补阳还五汤关键成分3个。GO分析结果显示,补阳还五汤治疗动脉粥样硬化潜在生物学过程有炎症反应、药物反应等43个。KEGG通路富集分析结果显示,补阳还五汤抗动脉粥样硬化通路主要涉及到Toll样受体信号通路、T细胞受体信号传导途径、核转录因子-kappaB(NF-κB)信号通路等。结论:补阳还五汤主要通过红花、黄芪所含有的3个关键成分槲皮素、木犀草素和山奈酚,以及Toll样受体信号通路、NF-κB信号通路、T细胞受体信号通路发挥抗炎症反应机制,达到抗动脉粥样硬化的作用。

Abstract:
Objective:To investigate the mechanism of action of the anti-atherosclerotic effect of Buyang Huanwu decoction based on network pharmacology. Methods:The TCMSP database was used to search for the constituents and targets of the traditional Chinese medicine,and SwissTargetPrediction and PubChem online websites were used to predict the targets of the constituents. OMIM,DisGeNE,TTD,and DrugBank databases were used to search for the targets of atherosclerosis. The Venn diagram was used to show the intersection between the targets of Buyang Huanwu decoction and the targets of atherosclerosis. STRING network was used to construct a protein-protein interaction network and screen out key targets. Cytoscape 3.6.0 was used to screen out the main constituents of Buyang Huanwu decoction,and DAVID6.8 website was used to perform GO functional analysis and KEGG pathway enrichment analysis of the intersecting targets. Results:There were 94 intersecting targets of Buyang Huanwu decoction and atherosclerosis,and Buyang Huanwu decoction had 62 active constituents and 3 key constituents. The GO analysis showed that Buyang Huanwu decoction had 43 potential biological processes in the treatment of atherosclerosis,including inflammatory response and drug reaction. The KEGG pathway enrichment analysis showed that the Toll-like receptor signaling pathway,the T cell receptor signaling pathway,and the nuclear factor-kappa B (NF-κB) signaling pathway were mainly involved in the treatment of atherosclerosis with Buyang Huanwu decoction. Conclusion:Buyang Huanwu decoction exerts an anti-atherosclerotic effect through the anti-inflammatory response mechanism via the three key constituents quercetin,luteolin,and kaempferol in Carthamus tinctorius and Astragalus membranaceus and the pathways including the Toll-like receptor signaling pathway,the NF-κB signaling pathway,and the T cell receptor signaling pathway.
