基于网络药理学探讨玉屏风散治疗支气管哮喘的作用机制

摘要:
目的:运用网络药理学筛选玉屏风散治疗支气管哮喘的作用靶标及信号通路,为临床治疗提供有力依据。方法:利用TCMSP筛选中药活性成分和有效成分靶点信息。利用Uniprot数据库和Excel找出有效成分靶点信息基因名。在GeneCards、OMIM、Drugbank数据库搜集疾病相关靶标,采用Venny软件对中药靶点基因与疾病靶点基因取交集,得到Venny交集图。将各个中药靶点基因共同输入Cytoscape软件,得到中药-靶标调控网络。通过String软件构建中药-疾病共同靶标的蛋白互作网络。将中药-疾病共同靶点基因进行GO和KEGG功能富集分析,采用Cytoscape软件做出共同靶点通路。结果:筛选得到玉屏风散治疗支气管哮喘的主要活性成分17个,中药-疾病共同靶点基因114个,推断其作用机制可能与糖尿病并发症中的AGE-RAGE信号通路、癌症途径、IL-17信号通路、PI3K-Akt信号通路等相关,并且可能起到关键作用。结论:基于网络药理学探讨玉屏风散治疗支气管哮喘的作用机制,有利于临床治疗。

Abstract:
Objective:To investigate the targets and signaling pathways of Yupingfeng powder in the treatment of bronchial asthma based on network pharmacology,and to provide a basis for clinical treatment.Methods:TCMSP database was used to screen out the active components and targets of Yupingfeng powder,and Uniprot database and Excel were used to find the gene names of these targets.GeneCards,OMIM,and Drugbank databases were searched for disease-related targets,and Venny software was used to obtain the Venny diagram of traditional Chinese medicine (TCM) target genes and disease target genes.TCM target genes were entered into Cytoscape software to obtain the TCM-target regulatory network,and String software was used to construct a protein-protein interaction network of TCM-disease targets.GO and KEGG functional enrichment analyses were performed for the common TCM-disease target genes,and Cytoscape software was used to find common target pathways.Results:A total of 17 active components of Yupingfeng powder and 114 common TCM-disease targets were screened out for the treatment of bronchial asthma,and its mechanism of action might be associated with the AGE-RAGE signaling pathway in diabetic complication,the cancer pathway,the IL-17 signaling pathway,and the PI3K-Akt signaling pathway.Conclusion:The mechanism of action of Yupingfeng powder in the treatment of bronchial asthma based on network pharmacology may help with clinical treatment.
