目的:通过网络药理学方法建立六味地黄丸“活性成分-靶点-通路”图,初步探讨六味地黄丸治疗糖尿病肾病的药理机制。方法:通过TCMSP获取六味地黄丸各单药的活性成分和潜在靶点,运用Genecard筛选得分≥20分的糖尿病肾病相关靶点,STRING构建蛋白相互作用网络,Cytoscape拓扑分析网络图,R语言进行GO和KEGG通路富集分析。结果:网络分析得出41个活性成分,共涉及21个靶点和63条通路。其中槲皮素和山奈酚是主要化合物,关键靶点AKT、VEGFA、NOS3、IL6、SERPINE1富集在重要通路AGE-RAGE信号中。结论:为进一步研究六味地黄丸治疗糖尿病肾病的潜在活性成分及其可能的作用机制提供了思路和理论基础。 |

Objective:To investigate the pharmacological mechanism of Liuwei Dihuang pills in the treatment of diabetic nephropathy by establishing the “active component-target-pathway” network of Liuwei Dihuang pills based on network pharmacology.Methods:Traditional Chinese Medicine Systems Pharmacology was used to obtain the active components and potential targets of Liuwei Dihuang pills,and Genecard was used to screen out the targets of diabetic nephropathy with a score of ≥20.STRING was used to establish a protein-protein interaction network,Cytoscape topology was used to analyze the network,and R language was used to perform gene ontology analysis and KEGG pathway enrichment analysis.Results:A total of 41 active components were obtained by the network analysis,which involved 21 targets and 63 pathways.Quercetin and kaempferol were the main components,and the key targets AKT,VEGFA,NOS3,IL6,and SERPINE1 were enriched in the important AGE-RAGE signaling pathway.Conclusion:This study provides ideas and a theoretical basis for further research on the potential active components and possible mechanism of action of Liuwei Dihuang pills in the treatment of diabetic nephropathy. |
