心痛泰颗粒对动脉粥样硬化小鼠血脂及VCAM-1、TNF-α的影响

摘要:
目的:探讨心痛泰颗粒对动脉粥样硬化(AS)小鼠血脂及VCAM-1、TNF-α的影响。方法:将10只野生型小鼠予以普通饲料喂养作为空白对照组,将40只ApoE基因敲除(ApoE-/-)小鼠予以高脂饲料喂养造模,模型制备成功后,将其随机分为模型组、心痛泰低剂量组、心痛泰高剂量组和他汀组,每组各10只,分别予以0.9%氯化钠注射液、心痛泰低剂量、心痛泰高剂量及阿托伐他汀钙片进行胃内给药,连续用药8周。检测各组小鼠用药前后的血清总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)及血管细胞黏附分子(VCAM)-1、肿瘤坏死因子-α(TNF-α)的水平变化。结果:与模型组、空白对照组比较,心痛泰低、高剂量组及他汀组可明显降低TC、TG、LDL-C浓度,升高HDL-C血清浓度,控制VCAM-1、TNF-α浓度并减少其阳性表达率,差异均有统计学意义(P<0.05);其中,心痛泰高剂量组、他汀组疗效优于心痛泰低剂量组,差异有统计学意义(P<0.05),他汀组与心痛泰高剂量组在降脂疗效方面比较,差异无统计学意义(P>0.05),在降低VCAM-1、TNF-α浓度方面,他汀组优于心痛泰高剂量组(P<0.05)。结论:心痛泰能够调节血脂、控制炎症,调节VCAM-1、TNF-α的阳性表达,从而干预动脉内斑块的形成及发展,起到预防AS的作用。

Effect of Xintongtai granules on blood lipids,vascular cell adhesion molecule-1,and tumor necrosis factor-α in mice with atherosclerosis

Abstract:
Objective:To investigate the effect of Xintongtai granules on blood lipids,vascular cell adhesion molecule-1 (VCAM-1),and tumor necrosis factor-α (TNF-α) in mice with atherosclerosis (AS).Methods:Ten wild-type mice were given normal diet and were selected as blank control group.A total of 40 mice with apolipoprotein E gene knockout were given high-fat diet to establish a model of AS,and after the model was successfully established,these mice were randomly divided into model group,low- and high-dose Xintongtai groups,and statin group,with 10 mice in each group.The mice in the model group were given intragastric administration of 0.9% sodium chloride injection,those in the low- and high-dose Xintongtai groups were given intragastric administration of low- and high-dose Xintongtai,and those in the statin group were given intragastric administration of atorvastatin calcium tablets.The course of treatment was 8 weeks for all group.Serum levels of total cholesterol (TC),triglyceride (TG),low-density lipoprotein cholesterol (LDL-C),high-density lipoprotein cholesterol (HDL-C),VCAM-1,and TNF-α were measured before and after administration.Results:Compared with the model group and the blank control group,the high- and low-dose Xintongtai groups and the statin group had significant reductions in TC,TG,and LDL-C and a significant increase in serum HDL-C,with controlled concentrations and reduced positive rates of VCAM-1 and TNF-α (P<0.05).The high-dose Xintongtai group and the statin group had a significantly better treatment outcome than the low-dose Xintongtai group (P<0.05),while there was no significant difference in lipid-lowering efficacy between the statin group and the high-dose Xintongtai group (P>0.05).Compared with the high-dose Xintongtai group,the statin group had significantly better reductions in the concentrations of VCAM-1 and TNF-α (P<0.05).Conclusion:Xintongtai exerts an intervention effect on the formation and development of arterial plaques and helps to prevent AS by regulating blood lipids,controlling inflammation,and regulating the expression of VCAM-1 and TNF-α.
