龙虎交战针法对膝骨关节炎大鼠丝裂原活化蛋白激酶信号通道的影响

摘要:
目的:观察龙虎交战针法对膝骨关节炎家兔关节软骨组织中丝裂原活化蛋白激酶(MAPKs)信号通路蛋白表达的影响,以探讨其作用机制。方法:建立膝骨关节炎大鼠模型,随机分为正常组、模型组、普通针刺组和龙虎交战组。正常组和模型组均常规饲养,不予治疗。普通针刺组和龙虎交战组分别采用常规针刺法和龙虎交战针法治疗。对各组大鼠进行行为学评分;取大鼠膝关节软骨组织,观察其病理形态变化;采用免疫组化观察软骨组织中丝裂原活化蛋白激酶(MAPKs)信号通路中ERK、P38MAPK、JNK蛋白的表达。结果:与正常组比较,模型组大鼠行为学评分降低,JNK、ERK、P38蛋白活性的表达升高(P<0.05,P<0.01);与模型组比较,普通针刺组、龙虎交战组大鼠行为学评分升高,JNK、ERK、P38蛋白表达降低(P<0.05,P<0.01),且龙虎交战组优于普通针刺组(P<0.05,P<0.01)。结论:龙虎交战针法能有效地调节MAPKs信号通路中ERK、P38MAPK、JNK蛋白活性的表达,降低MAPKs活性,从而有效缓解关节疼痛,促进关节软骨修复。这可能是龙虎交战针法治疗膝骨关节炎起效的作用机制之一。

Abstract:
Objective:To investigate the effect of dragon-tiger contending acupuncture on the mitogen-activated protein kinase(MAPK) signaling pathway in articular cartilage in rats with knee osteoarthritis and its mechanism of action.Methods:A rat model of knee osteoarthritis was established.The rats were randomly divided into normal group,model group,conventional acupuncture group,and dragon-tiger contending group.The rats in the normal group and the model group were given routine feeding without any treatment,and those in the conventional acupuncture group and the dragon-tiger contending group were given conventional acupuncture or dragon-tiger contending acupuncture.Behavioral score was determined for all rats;knee articular cartilage was collected to observe its pathomorphological changes;immunohistochemistry was used to measure the expression of ERK,P38MAPK,and JNK proteins involved in the MAPK signaling pathway in cartilage.Results:Compared with the normal group,the model group had a significant reduction in behavioral score and significant increases in the expression of JNK,ERK,and P38 proteins(P<0.05 or P<0.01).Compared with the model group,the conventional acupuncture group and the dragon-tiger contending group had a significant increase in behavioral score and significant reductions in the expression of JNK,ERK,and P38 proteins(P<0.05 or P<0.01),and the dragon-tiger contending group had better improvements than the conventional acupuncture group(P<0.05 or P<0.01).Conclusion:Dragon-tiger contending acupuncture can effectively regulate the expression of ERK,P38MAPK,and JNK proteins in the MAPK signaling pathway,reduce the activity of MAPK,and thus alleviate joint pain and promote the repair of articular cartilage,which may be one of the mechanisms of action of dragon-tiger contending acupuncture in the treatment of knee osteoarthritis.
