基于网络药理学探讨薏苡附子败酱汤合四逆散治疗难治性面部痤疮的作用机制

摘要:
目的:分析王清坚教授基于“十二纲-病理-方证”辨证体系运用薏苡附子败酱汤合四逆散治疗难治性面部痤疮的作用机制。方法:运用网络药理学方法,借助TCMSP数据库获取薏苡附子败酱汤合四逆散的活性成分及相关靶点,使用GeneCards和OMIM数据库检索得到难治性面部痤疮的作用靶点;通过R语言将药物有效成分靶点和疾病靶点取交集,并绘制韦恩图,运用STRING数据库对交集靶点进行蛋白质-蛋白质相互作用(PPI)网络分析;采用Cytoscape软件构建薏苡附子败酱汤合四逆散治疗难治性面部痤疮的“药物-活性成分-疾病靶点”网络,并筛选出核心活性成分及靶点;利用DAVID数据库进行基因本体(GO)功能和京都基因与基因组百科全书(KEGG)通路富集分析。结果:共获得169种药物活性成分,对应2781个作用靶点,其中与难治性面部痤疮的共同靶点有77个。PPI网络筛选得到AP-1转录因子亚基、转录因子p65、核因子κB抑制因子α等9个核心靶点;GO功能富集分析得到条目共1878条,KEGG通路富集分析显示其主要涉及血脂和动脉粥样硬化、化学致癌-受体激活、肿瘤坏死因子、白细胞介素-17和核转录因子-κB等信号通路。结论:薏苡附子败酱汤合四逆散可多成分、多靶点、多通路地治疗难治性面部痤疮,为进一步发掘其潜在作用机制提供了新思路。

Abstract:
Objective: To investigate the mechanism of action of Yiyi Fuzi Baijiang decoction combined with Sini powder in the treatment of refractory facial acne by Professor Wang Qingjian based on the “twelve principles-pathology-prescription/syndrome” syndrome differentiation system.Methods: Based on the methods of network pharmacology,TCMSP database was used to obtain the active components of Yiyi Fuzi Baijiang decoction combined with Sini powder and related targets,and GeneCards and OMIM databases were used to obtain the action targets of refractory facial acne.R language was used to obtain the intersecting targets between the targets of effective constituents of drugs and the targets of the disease and plot the Venn diagram,and STRING database was used to perform a protein-protein interaction (PPI) analysis of the intersecting targets.Cytoscape software was used to construct a drug-active component-disease target network for Yiyi Fuzi Baijiang decoction combined with Sini powder in the treatment of refractory facial acne and obtain the core active components and targets,and DAVID database was used to perform gene ontology (GO) functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis.Results: A total of 169 active components of the drugs were obtained,corresponding to 2781 action targets,among which there were 77 common targets with refractory facial acne.The PPI network analysis obtained 9 core targets including AP-1transcription factor subunit,transcription factor p65,and nuclear factor kappa B repressor.GO functional enrichment analysis obtained 1878 terms,and KEGG pathway enrichment analysis showed that they mainly involved the signaling pathways such as blood lipids and atherosclerosis,chemical carcinogenesis-receptor activation,tumor necrosis factor,interleukin-17,and nuclear factor-κB.Conclusion: Yiyi Fuzi Baijiang decoction combined with Sini powder exerts a therapeutic effect on refractory facial acne through multiple components,targets,and pathways,which provides new ideas for further exploring its potential mechanism of action.
