基于网络药理学探讨赤芍抗血吸虫病及肝纤维化的作用机制

摘要:
目的:通过网络药理学方法探讨赤芍有效成分治疗血吸虫病及肝纤维化的潜在作用机制。方法:通过TCMSP数据库检索并收集赤芍有效成分及其对应的作用靶点,借助UniProt数据库将靶点蛋白的genename转换为genesymbol;通过GeneCards、OMIM、TTD疾病数据库搜集血吸虫病及肝纤维化相关靶点;将赤芍成分靶点和疾病靶点取交集,获得赤芍治疗血吸虫病及肝纤维化的潜在靶点,利用Cytoscape 3.8.2软件构建“药物-成分-靶点-疾病”和“成分-靶点-通路”网络;通过STRING数据库对交集靶点进行蛋白质-蛋白质相互作用(PPI)网络分析,采用Cytoscape 3.8.2软件进行分析和可视化呈现;通过Metascape数据库进行基因本体(GO)功能、京都基因与基因组百科全书(KEGG)通路富集分析。结果:筛选得到赤芍活性成分29种,成分靶点与血吸虫病及肝纤维化疾病的交集靶点19个,其中肿瘤抑制蛋白(TP53)、前列腺素内过氧化物合酶(PTGS2)、肿瘤坏死因子(TNF)、基质金属蛋白酶9(MMP9)、白细胞介素-6(IL-6)、血管内皮生长因子A(VEGFA)、白细胞介素8(IL-8)、缺氧诱导因子1A(HIF1A)、转化生长因子B1(TGFB1)、基质金属蛋白酶2(MMP2)为核心靶点,涉及的主要信号通路为癌症信号通路、乙型肝炎、糖尿病并发症中的晚期糖基化终产物及其受体(AGE-RAGE)信号通路、动脉粥样硬化。结论:本研究初步预测了赤芍抗血吸虫病及肝纤维化的作用机制,可为今后药效物质基础研究提供参考。

Mechanism of action of Radix Paeoniae Rubra in treatment of schistosomiasis and liver fibrosis:A study based on network pharmacology

Abstract:
Objective:To investigate the potential mechanism of action of the effective constituents of Radix Paeoniae Rubra in the treatment of schistosomiasis and liver fibrosis based on network pharmacology.Methods:TCMSP database was used to search and obtain the effective constituents of Radix Paeoniae Rubra and their corresponding targets,and UniProt database was used to convert the gene name of target proteins into gene symbol.GeneCards,OMIM,and TTD disease targets were used to collect the targets associated with schistosomiasis and liver fibrosis.The targets of the constituents of Radix Paeoniae Rubra were intersected with the targets of the diseases to obtain the potential targets of Radix Paeoniae Rubra in the treatment of schistosomiasis and liver fibrosis,and Cytoscape 3.8.2 was used to construct the “drug-component-target-disease” network and the “component-target-pathway” network.STRING database was used to perform a protein-protein interaction (PPI) network analysis of the intersecting targets,and Cytoscape 3.8.2 was used for analysis and visualization.Metascape database was used to perform the gene ontology functional enrichment analysis and the Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis.Results:The above analyses obtained 29 active components of Radix Paeoniae Rubra and 19 intersecting targets between the targets of the components and the targets of schistosomiasis and liver fibrosis,among which tumor protein 53,prostaglandin endoperoxide synthase 2,tumor necrosis factor,matrix metalloproteinase-9,interleukin-6,vascular endothelial growth factor A,IL-8,hypoxia-inducible factor 1A,transforming growth factor B1,and matrix metalloproteinase-2 were core targets.The main signaling pathways involved were the cancer signaling pathway,the hepatitis B signaling pathway,the AGE-RAGE signaling pathway,and the atherosclerosis signaling pathway.Conclusion:This study preliminarily predicts the mechanism of action of Radix Paeoniae Rubra in the treatment of schistosomiasis and liver fibrosis,which provides a reference for subsequent studies on pharmacodynamic material basis.
