基于网络药理学探讨虎地肠溶胶囊治疗溃疡性结肠炎的作用机制

摘要:
目的:通过网络药理学探析虎地肠溶胶囊治疗溃疡性结肠炎(UC)的作用机制。方法:基于TCMSP和台湾中医药数据库筛查药物的潜在活性成分及靶点;采用GeneCards数据库获取UC的已知靶点,运用STRING数据库构建蛋白质-蛋白质相互作用(PPI)网络图;应用DAVID数据库对共同靶点基因进行基因本体(GO)功能和京都基因与基因组百科全书(KEGG)通路富集分析;通过Cytoscape 3.7.2软件和R语言软件对数据进行可视化。结果:虎地肠溶胶囊治疗UC的活性成分143种,关键成分包括山柰酚、β-谷甾醇、槲皮素等;潜在作用靶点69个,主要有肿瘤蛋白P53(TP53)、原癌基因蛋白(MYC)、胱天蛋白酶3(CASP3)、人雌激素受体α(ESR1)等靶基因。晚期糖基化产物-晚期糖基化终末产物受体(AGE-RAGE)信号通路、肿瘤坏死因子(TNF)信号通路。GO功能富集分析结果显示主要涉及DNA转录调控、RNA聚合酶Ⅱ特异性、核受体调控、单加氧酶活性、氧化还原反应等。KEGG通路富集分析结果显示主要涉及疾病相关信号通路、炎症相关信号通路和细胞凋亡信号通路。结论:虎地肠溶胶囊的多种活性成分具有从多靶点、多通路治疗UC的作用,为临床研究提供新思路。

Abstract:
Objective:To investigate the mechanism of action of Hudi enteric-coated capsules in the treatment of ulcerative colitis (UC) based on network pharmacology.Methods:TCMSP and Traditional Chinese Medicine Dadabase@Taiwan were used to screen for the potential active components and targets of the drug;GeneCards database was used to obtain the known targets of UC,and STRING database was used to construct a protein-protein interaction network;DAVID database was used to perform gene ontology (GO) functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis of common target genes;Cytoscape 3.7.2 and R language were used for data visualization.Results:A total of 143 active components were obtained for Hudi enteric-coated capsules,with the key components including kaempferol,beta-sitosterol,and quercetin,and 69 potential action targets were obtained,mainly TP53,MYC,CASP3,and ESR1.The AGE-RAGE signaling pathway and the tumor necrosis factor signaling pathway were important signaling pathways.The GO functional enrichment analysis showed the involvement of DNA transcription regulation,RNA polymerase II specificity,nuclear receptor regulation,monooxygenase activity,and redox reaction.The KEGG pathway enrichment analysis showed the involvement of the disease-related signaling pathway,inflammation-related signaling pathway,and the cell apoptosis signaling pathway.Conclusion:The multiple active components of Hudi enteric-coated capsules exert a therapeutic effect on UC through multiple targets and pathways,which provides new ideas for clinical research.
