电针对系统炎症诱导认知功能障碍小鼠海马CD137、CD137L及TREM2的影响

摘要:
目的:探讨电针足三里对脂多糖引起的系统炎症诱导认知功能障碍小鼠海马白细胞分化抗原137(CD137)、CD137配体(CD137L)和髓样细胞触发受体2(TREM2)的影响。 方法:将24只小鼠随机分为对照组、模型组和电针组,每组各8只,模型组、电针组腹腔注射脂多糖(1 mg/kg)进行造模,对照组给予同等容量0.9%氯化钠注射液。电针组于造模后及造模后24 h给予15 min 10 Hz、0.5 mA的连续波电刺激足三里穴,对照组、模型组不做处理。干预后采用新物体识别实验检测小鼠新物体识别指数以评价小鼠学习记忆能力,采用酶联免疫吸附法检测小鼠海马 CD137、CD137L和TREM2的含量。结果:与对照组比较,模型组小鼠新物体识别指数、TREM2含量显著降低,CD137、CD137L 含量显著升高(均P<0.01);与模型组比较,电针组小鼠新物体识别指数、TREM2含量显著升高,CD137、CD137L含量显著降低(均P<0.01)。结论:电针足三里可改善系统炎症诱导的认知功能障碍,其作用机制可能与降低海马区CD137、CD137L含量及提高TREM2含量有关。

Effect of electroacupuncture on hippocampal cluster of differentiation 137,cluster of differentiation 137 ligand,and triggering receptor expressed on myeloid cells 2 in mice with cognitive impairment induced by systemic inflammation

Abstract:
Objective:To investigate the effect of electroacupuncture at Zusanli on hippocampal cluster of differentiation 137 (CD137),CD137 ligand (CD137L),and triggering receptor expressed on myeloid cells 2 (TREM2) in mice with cognitive impairment induced by systemic inflammation due to lipopolysaccharide.Methods:A total of 24 mice were randomly divided into control group,model group,and electroacupuncture group,with 8 mice in each group.The mice in the model group and the electroacupuncture group were given intraperitoneal injection of lipopolysaccharide (1 mg/kg) for modeling,and those in the control group were given an equal volume of 0.9% sodium chloride injection.Immediately and at 24 hours after modeling,the mice in the electroacupuncture group were given continuous-wave electrical stimulation (10 Hz,0.5 mA) at Zusanli for 15 minutes,and those in the control group and the model group were not given any treatment.After intervention,the novel object recognition test was used to measure novel object recognition index to evaluate learning and memory abilities,and ELISA was used to measure the content of CD137,CD137L,and TREM2 in the hippocampus.Results:Compared with the control group,the model group had significant reductions in novel object recognition index and the content of TREM2 and significant increases in the content of CD137 and CD137L (all P<0.01),and compared with the model group,the electroacupuncture group had significant increases in novel object recognition index and the content of TREM2 and significant reductions in the content of CD137 and CD137L (all P<0.01).Conclusion:Electroacupuncture at Zusanli can improve cognitive impairment induced by systemic inflammation,possibly by reducing the content of CD137 and CD137L and increasing the content of TREM2 in the hippocampus.
