目的:探讨血根碱(SAN)对鼻咽癌细胞(CNE2细胞系)增殖和迁移的影响及其可能的作用机制。方法:采用MTT法检测SAN对CNE2细胞增殖的影响,采用细胞划痕实验研究SAN对CNE2细胞迁移能力的影响,采用Western Blot技术检测SAN对CNE2细胞增殖细胞核抗原(PCNA)和上皮-间质转化(EMT)相关蛋白E-钙黏蛋白(E-Cadherin)、N-钙黏蛋白(N-Cadherin)、波形蛋白(Vimentin)表达水平的影响。结果:MTT法检测结果显示,与阴性对照组比较,SAN可以抑制鼻咽癌CNE2细胞的增殖,而且SAN浓度越高,其抑制效应越显著(P<0.05)。细胞划痕实验结果显示,与溶剂对照组比较,SAN可以抑制鼻咽癌CNE2细胞迁移,而抑制效应与药物浓度相关(P<0.05)。Western Blot检测结果显示,与溶剂对照组比较,SAN能降低CNE2细胞中增殖相关蛋白PCNA和EMT相关蛋白N-Cadherin、Vimentin的表达水平,提高E-Cadherin的表达水平(P<0.05)。结论:血根碱能抑制鼻咽癌细胞增殖和迁移,可作为治疗鼻咽癌的潜在药物开展进一步研究。 |

Abstract: |
Objective:To investigate the effect of sanguinarine (SAN) on the proliferation and migration of nasopharyngeal carcinoma CNE2 cells and its possible mechanism of action.Methods:MTT assay was used to observe the effect of SAN on the proliferation of CNE2 cells,wound healing assay was used to observe the effect of SAN on the migration of CNE2 cells,and Western blot was used to observe the effect of SAN on the expression levels of proliferating cell nuclear antigen (PCNA) and epithelial-mesenchymal transition (EMT)-related proteins (E-cadherin,N-cadherin,and vimentin).Results:MTT assay showed that compared with the negative control group,SAN inhibited the proliferation of CNE2 cells in a concentration-dependent manner (P<0.05).Wound healing assay showed that compared with the solvent control group,SAN inhibited the migration of CNE2 cells in a concentration-dependent manner (P<0.05).Western blot showed that compared with the solvent control group,SAN reduced the expression levels of the proliferation-related protein PCNA and the EMT-related proteins N-cadherin and vimentin and increased the expression level of the EMT-related protein E-cadherin (P<0.05).Conclusion:SAN can inhibit the proliferation and migration of nasopharyngeal carcinoma cells and can thus be further studied as a potential therapeutic drug for nasopharyngeal carcinoma. |
