基于网络药理学探讨六味地黄丸合缩泉丸治疗尿崩症的作用机制

摘要:
目的:基于网络药理学探讨六味地黄丸合缩泉丸治疗尿崩症(DI)的作用机制。方法:利用TCMSP和 UniProt 数据库获取六味地黄丸合缩泉丸的有效成分和作用靶点,通过GeneCards、OMIM、DrugBank数据库预测疾病靶点,将有效成分靶点与疾病靶点取交集,构建韦恩图获取共同靶点,运用STRING数据库和Cytoscape 3.8.2软件绘制交集靶点的蛋白质-蛋白质相互作用(PPI)网络,经拓扑分析得到关键靶点,并进行基因本体(GO)功能和京都基因与基因组百科全书(KEGG)通路富集分析。结果:共获得六味地黄丸合缩泉丸有效成分 31 个,作用于 185 个靶点,DI靶点共 897 个,六味地黄丸合缩泉丸与DI交集靶点 39 个,主要有效成分有槲皮素、山柰酚、β-谷甾醇、薯蓣皂苷元等,关键靶点包括丝氨酸/苏氨酸蛋白激酶1(AKT1)、白细胞介素-1β(IL-1β)、基质金属蛋白酶(MMP9)、过氧化物酶体增生激活受体(PPARG)、趋化因子(CCL2)、肿瘤蛋白TP53(p53)等。GO功能富集得到1667个条目,包括生物过程1582个、分子功能78个、细胞组分7个;KEGG通路富集得到110条通路,主要涉及动脉粥样硬化信号通路、糖尿病并发症中的AGE-RAGE信号通路、疟疾、肿瘤坏死因子信号通路等。结论:本研究初步揭示了六味地黄丸合缩泉丸治疗DI的作用机制,并通过数据分析侧面印证了中医辨证论治理论体系的正确性,可为进一步研究提供借鉴。

Mechanism of action of Liuwei Dihuang pills combined with Suoquan pills in treatment of diabetes insipidus:A study based on network pharmacology

Abstract:
Objective:To investigate the mechanism of action of Liuwei Dihuang pills combined with Suoquan pills in the treatment of diabetes insipidus (DI) based on network pharmacology.Methods:TCMSP and UniProt databases were used to obtain the effective constituents and action targets of Liuwei Dihuang pills combined with Suoquan pills,and GeneCards,OMIM,and DrugBank databases were used to predict the disease targets of DI.Intersecting targets were obtained for the targets of the effective constituents and the disease targets,and a Venn diagram was plotted to obtain common targets.STRING database and Cytoscape 3.8.2 software were used to construct a protein-protein interaction network for the intersecting targets,and a topological analysis was performed to obtain the key targets.Gene ontology (GO) functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis were also performed.Results:A total of 31 effective constituents were obtained for Liuwei Dihuang pills combined with Suoquan pills,which acted on 185 targets.A total of 897 targets were obtained for DI,and 39 intersecting targets were obtained for Liuwei Dihuang pills combined with Suoquan pills and DI.The main effective constituents included quercetin,kaempferol,beta-sitosterol,and dioscin,and the key targets included AKT1,IL-1β,MMP9,PPARG,CCL2,and p53.The GO functional enrichment analysis obtained 1667 items,including 1582 biological processes,78 molecular functions,and 7 cellular components,and the KEGG pathway enrichment analysis obtained 110 pathways,which mainly involved the atherosclerosis signaling pathway,the AGE-RAGE signaling pathway in diabetic complication,malaria,and the tumor necrosis factor signaling pathway.Conclusion:This study preliminarily reveals the mechanism of action of Liuwei Dihuang pills combined with Suoquan pills in the treatment of DI and confirms the correctness of the theoretical system of TCM syndrome differentiation-based treatment through data analysis,which provides a reference for further research.
