苍附导痰汤加减对肥胖型多囊卵巢综合征大鼠的干预作用研究

摘要:
目的:探讨苍附导痰汤加减对肥胖型多囊卵巢综合征(PCOS)大鼠内分泌、糖脂代谢及核因子κB(NF-κB) p65蛋白的影响。方法:将40只大鼠随机分为空白组(10只)和造模组(30只)。空白组予普通饲料,造模组采用来曲唑联合高脂高糖饮食建立肥胖型PCOS大鼠模型。将30只PCOS大鼠随机分为模型组、西药组、中药组,每组各10只。空白组和模型组给予等体积0.9%氯化钠注射液,西药组予以二甲双胍(135 mg/kg·d),中药组予以苍附导痰汤加减(14.49 g/kg·d),连续灌胃干预21 d。给药结束后检测各组大鼠的体质量、子宫与卵巢指数、血清卵泡刺激素(FSH)、促黄体生成素(LH)、总睾酮(T)、空腹血糖(FBG)、胰岛素抵抗指数(HOMA-IR)、空腹胰岛素(FINS)、总胆固醇(TC)、三酰甘油(TG)水平及卵巢组织NF-κB p65蛋白表达。结果:模型组、西药组、中药组治疗前后的体质量及治疗后的FSH、LH、T水平,模型组的子宫指数及HOMA-IR、FINS、TC、TG、NF-κB p65蛋白表达水平,与空白组比较,差异均有统计学意义(P<0.05或P<0.01)。西药组、中药组的子宫指数及FSH、T、HOMA-IR、FINS、TG、NF-κB p65蛋白表达水平,西药组的LH水平,与模型组比较,差异均有统计学意义(P<0.05或P<0.01)。结论:苍附导痰汤加减能有效改善肥胖型PCOS大鼠内分泌激素及糖脂代谢水平、胰岛素抵抗,其作用机制可能与NF-κB信号通路相关。

Intervention effect of modified Cangfu Daotan decoction on rats with obese polycystic ovary syndrome

Abstract:
Objective:To investigate the effect of modified Cangfu Daotan decoction on endocrine,glucose and lipid metabolism,and nuclear factor-kappa B (NF-κB) in a rat model of obese polycystic ovary syndrome (PCOS).Methods:A total of 40 rats were randomly divided into blank group with 10 rats and modeling group with 30 rats.The rats in the blank group were given normal diet,and those in the modeling group were given letrozole combined with high-fat high-glucose diet to establish a rat model of obese PCOS.The 30 PCOS rats were randomly divided into model group,Western medicine group,and traditional Chinese medicine (TCM) group,with 10 rats in each group.The rats in the blank group and the model group were given an equal volume of 0.9% sodium chloride,those in the Western medicine group were given metformin (135 mg/kg·d),and those in the TCM group were given modified Cangfu Daotan decoction (14.49 g/kg·d),by gavage for 21 consecutive days.Related indices were measured after administration,including body weight,uterus index and ovary index,the serum levels of follicle-stimulating hormone (FSH),luteinizing hormone (LH),total testosterone (T),fasting blood glucose (FBG),Homeostasis Model Assessment of Insulin Resistance (HOMA-IR) index,fasting insulin (FINS),total cholesterol (TC),and triglyceride (TG),and the expression of NF-κB p65 protein in ovary tissue.Results:There were significant differences in body weight before and after treatment and the levels of FSH,LH,and T after treatment between the model group/Western medicine group/TCM group and the blank group,and there were significant differences in uterus index,HOMA-IR,FINS,TC,TG,and NF-κB p65 protein expression between the model group and the blank group (P<0.05 or P<0.01).There were significant differences in uterus index,FSH,T,HOMA-IR,FINS,TG,and NF-κB p65 protein expression between the Western medicine group/TCM group and the model group,and there was a significantly difference in LH between the Western medicine group and the model group (P<0.05 or P<0.01).Conclusion:Modified Cangfu Daotan decoction can effectively improve endocrine hormones,glucose and lipid metabolism,and insulin resistance in rats with obese PCOS,which may be associated with the NF-κB signaling pathway.
