基于网络药理学探讨黄连防治2型糖尿病的作用机制

摘要:
目的:运用网络药理学分析黄连防治2型糖尿病的作用机制。方法:通过TCMSP及UniProt数据库查询黄连主要活性成分与相关作用靶点,在GeneCards、OMIM数据库中检索2型糖尿病靶点,并通过OmicShare软件获得黄连与2型糖尿病共同靶点,运用String数据库对共同靶点进行蛋白质相互作用(PPI)分析,构建黄连防治2型糖尿病靶点蛋白互作(PPI)网络,利用CytoScape软件构建黄连-化合物-靶点-2型糖尿病网络图,并通过R语言软件对其防治2型糖尿病作用靶点的基因本体论(GO)、京都基因与基因组百科全书(KEGG)通路进行富集分析。结果:获得黄连成分中具有类药性、口服吸收良好的化合物10种,其防治2型糖尿病的潜在作用靶点94个,包括丝氨酸、苏氨酸蛋白激酶1(AKT1)、血管内皮生长因子A(VEGFA)、白细胞介素6(IL-6)、丝裂原活化蛋白激酶(CASP3)、表皮生长因子(EGF)、转录因子AP-1(JUN)等,信号通路主要有缺氧诱导因子-1(HIF-1)、丝裂原活化蛋白激酶(MAPK)、脂酰肌醇3激酶-蛋白激酶B(PI3K/AKT)等。结论:黄连可通过多组分、多靶点、多机制防治2型糖尿病,为深入研究黄连防治2型糖尿病的潜在作用机制提供理论基础。

Abstract:
Objective:To investigate the mechanism of action of Coptis chinensis in the prevention and treatment of type 2 diabetes based on network pharmacology.Methods:TCMSP and UniProt databases were used to obtain the main active components of Coptis chinensis and related action targets,and GeneCards and OMIM databases were searched to obtain the targets of type 2 diabetes.OmicShare software was used to obtain the common targets of Coptis chinensis and type 2 diabetes,and String database was used to perform a protein-protein interaction (PPI) analysis of the common targets and then construct a PPI network for Coptis chinensis in the prevention and treatment of type 2 diabetes.CytoScape software was used to construct a Coptis chinensis-compound-target-type 2 diabetes network,and R language software was used to perform gene ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analyses for the action targets of Coptis chinensis in the prevention and treatment of type 2 diabetes.Results:A total of 10 compounds with drug-likeness and good oral absorption were screened out from the components of Coptis chinensis,with 94 potential action targets in the prevention and treatment of type 2 diabetes,including AKT1,vascular endothelial growth factor A,interleukin-6,CASP3,epidermal growth factor,and transcription factor AP-1 (JUN),and the main signaling pathways included hypoxia-inducible factor-1,mitogen-activated protein kinase,and phosphatidylinositol 3-kinase/protein kinase B.Conclusion:Coptis chinensis can prevent and treat type 2 diabetes through multiple components,targets,and mechanisms,which provides a theoretical basis for in-depth research on the potential mechanism of action of Coptis chinensis in the prevention and treatment of type 2 diabetes.
