丹荔输通汤对慢性输卵管炎模型大鼠干预作用的实验研究

摘要:
目的:观察丹荔输通汤对慢性输卵管炎模型大鼠的干预作用。方法:84只SD大鼠按体质量编号,随机抽取11只为空白组,余73只采用苯酚明胶造模法制备输卵管炎性阻塞性模型。将造模成功的66只实验大鼠随机分为6组,即模型组、中药高剂量灌胃+灌肠组(中药高剂量组)、中药中剂量灌胃+灌肠组(中药中剂量组)、中药低剂量灌胃+灌肠组(中药低剂量组)、中药中剂量灌胃组(中药灌胃组)、西药对照组(西药组),每组各11只。模型组灌胃等容积0.9%氯化钠注射液,中药高、中、低剂量组及中药灌胃组分别给予灌胃、灌肠丹荔输通汤,对照组灌胃阿奇霉素混悬液与等容积0.9%氯化钠注射液。连续干预30d。观察各组大鼠的输卵管形态、组织病理学及肿瘤坏死因子(TNF-α)、白介素-6(IL-6)水平。结果:干预后,肉眼观察发现,与模型组相比,各用药组输卵管组织形态可见不同程度的改善。组织病理学结果显示,中药高剂量组输卵管形态及功能基本正常,中药中剂量组、中药灌胃组、西药组输卵管轻度炎症表现。与空白组比较,模型组TNF-α、IL-6表达升高,与模型组比较,各给药组TNF-α、IL-6表达均降低,差异有统计学意义(P<0.01)。与西药组比较,中药高、中剂量组TNF-α、IL-6表达降低(P<0.05);中药高、中剂量组TNF-α、IL-6水平低于中药低剂量组、中药灌胃组,中药中剂量组TNF-α、IL-6水平低于中药灌胃组,差异均有统计学意义(P<0.01)。结论:丹荔输通汤治疗慢性输卵管炎可能通过调节TNF-α、IL-6而抑制炎性反应,从而改善全身及局部组织炎症状态。

Intervention effect of Danli Shutong decoction on a rat model of chronic salpingitis:An experimental study

Abstract:
Objective:To investigate the intervention effect of Danli Shutong decoction on a rat model of chronic salpingitis.Methods:A total of 84 Sprague-Dawley rats were selected;according to the number of body weight,11 rats were randomly selected as blank group,and the remaining 73 rats were used to establish a model of chronic salpingitis by the phenol-gelatin modeling method.After successful modeling,66 experimental rats were randomly divided into model group,high-dose traditional Chinese medicine (TCM) intragastric administration+enema group (high-dose TCM group),middle-dose TCM intragastric administration+enema group (middle-dose TCM group),low-dose TCM intragastric administration+enema group (low-dose TCM group),middle-dose TCM intragastric administration group (TCM intragastric administration group),and Western medicine control group (Western medicine group),with 11 rats in each group.The rats in the model group were given an equal volume of 0.9% sodium chloride injection by gavage,and those in the high-,middle-,and low-dose TCM groups and the TCM intragastric administration group were given Danli Shutong decoction by gavage or enema with Danli Shutong decoction;the rats in the control group were given azithromycin suspension and an equal volume of 0.9% sodium chloride injection by gavage.Intervention was given for 30 consecutive days.All groups were observed in terms of fallopian tube morphology,histopathology,and levels of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6).Results:After intervention,observation with the naked eye showed that compared with the model group,the administration groups had varying degrees of improvement in fallopian tube histomorphology.Histopathological results showed that the high-dose TCM group had basically normal fallopian tube morphology and function,while the middle-dose TCM group,the TCM intragastric administration group,and the Western medicine group had mild inflammation in the fallopian tube.Compared with the blank group,the model group had significant increases in the expression of TNF-α and IL-6,and compared with the model group,the administration groups had significant reductions in the expression of TNF-α and IL-6 (P<0.01).Compared with the Western medicine group,the high- and middle-dose TCM groups had significant reductions in the expression of TNF-α and IL-6 (P<0.05);the high- and middle-dose TCM groups had significantly lower levels of TNF-α and IL-6 than the low-dose TCM group and the TCM intragastric administration group,and the middle-dose TCM group had significantly lower levels of TNF-α and IL-6 than the TCM intragastric administration group (P<0.01).Conclusion:In the treatment of chronic salpingitis,Danli Shutong decoction inhibits inflammatory response by regulating TNF-α and IL-6 and thus improves systemic and local inflammation.
