基于网络药理学探讨青风藤治疗糖尿病肾病的作用机制

摘要:
目的:运用网络药理学对青风藤治疗糖尿病肾病(DN)的作用机制进行研究。方法:通过TCMSP数据库筛查出青风藤的活性成分以及作用靶点,并在TTD、DrugBank、DisGeNet数据库中共同筛选出DN的基因靶点,通过Venny 2.1.0得出青风藤和DN基因靶点的交集,运用STRING 11.0数据库和Cytoscape 3.8.2软件制作蛋白互作网络图,运用Web Gestal数据库对相应基因靶点进行GO功能富集分析及KEGG通路富集分析。结果:共筛选出6种有效活性成分,潜在靶点蛋白58个,其中治疗DN有效活性成分4种,潜在作用靶点18个。GO功能以及KEGG通路富集分析发现,青风藤治疗DN与凋亡通路、AGE-RAGE信号通路相关,并通过调控这些通路降低尿蛋白的产生、保护细胞组织、延缓肾小球纤维化及肾脏硬化。结论:青风藤可以通过多靶点、多通路的协同作用,延缓DN的进展,可为临床用药提供依据。

Abstract:
Objective:To investigate the mechanism of action of Sinomenium acutum in the treatment of diabetic nephropathy based on network pharmacology.Methods:TCMSP database was used to obtain the active components and action targets of Sinomenium acutum,and TTD,DrugBank,and DisGeNet databases were used to obtain the gene targets of DN.Venny 2.1.0 was used to obtain the intersecting targets of Sinomenium acutum and the gene targets of DN;STRING 11.0 database and Cytoscape 3.8.2 software were used to plot a protein-protein interaction network;Web Gestal database was used to perform GO functional enrichment analysis and KEGG pathway enrichment analysis of corresponding gene targets.Results:A total of 6 effective components and 58 potential target proteins were screened out,among which there were 4 effective components and 18 potential action targets for the treatment of DN.GO functional enrichment analysis and KEGG pathway enrichment analysis showed that in the treatment of DN,Sinomenium acutum was associated with the apoptosis signaling pathway and the AGE-RAGE signaling pathway and reduced the production of urinary protein,protected cellular tissue,and delayed glomerular fibrosis and nephrosclerosis by regulating these pathways.Conclusion:Sinomenium acutum can delay the progression of DN through the synergistic effect of multiple targets and pathways,which provides a basis for clinical medication.
